Objective: To investigate JNK expression in esophageal squamous cell carcinoma(ESCC) and relationship with clinicopathological features, provide a theoretical basis of the prevention and treatment for a new clinical target.Methods: Immunohistochemistry SP method, reverse transcription of polymerase chain reaction (RT-PCR) and Western blot were employed , to examine JNK expression ESCC,the software SPSS13.0 was used to anlysis the result of ESCC .Results:1 RT-PCR results showed that Esophageal cancer JNK bands was higher than the density and width of the mucosa. JNK mRNA expression in esophageal cancer was significantly higher than in the mucosal tissues. The relative expression of ESCC JNK mRNA volume were (0.648±0.089), mucosal tissue (0.431±0.095)respectively.There was significant differences (P<0.05). JNK mRNA relative expression level in well and moderate–differentiated group was 0.627±0.071 , while that was significantly higher erpression in low-differentiated 0.731±0.063 ,Differences was statistically significant after statistical analysis (P <0.05) .Lymph node metastasis and non-lymph metastasis were the relative expression levels:0.623±0.086, 0.690±0.066 (P<0.05). The JNK mRNA expression was not correlated with depth of invasion, gender and age (P>0.05).2 Western-blot result show that JNK protein expression higher in carcinoma than in adjacent esophageal mucosa group(P<0.05);higher in Lymph node metastasis than in non-lymph metastasis(P<0.05);higher in low differentiation than in well and moderate differentiation(P <0.05);there was not correlated with depth of invasion, gender and age (P> 0.05).3 The immunohistochemical staining result:JNK protein expression higher in carcinoma than in adjacent esophageal mucosa group (χ~2= 39.846, P<0.01).3.1 The positive rates of expression of JNKin ESCC of invasive depth were submucosa 80.00%, muscular layer 87.5% and fibrous layer 94.90% respectively. The difference was significant among them (P<0.05).3.2 The positive rates of expression of JNK in ESCC of clinical stagesⅠ-ⅡandⅢ-Ⅳwere 88.9% (24/27),97.60%(32/33) respectively,The difference was significant among them (P<0.05).results showed that the positive correlation between the later clinical stages,JNK protein expression was increased.3.3 Lymph node metastasis and non-lymph metastasis JNK protein expression was 99.3% (31/33), 92.6% (25/27), statistically significant difference between the groups (χ~2= 12.903, P <0.05).3.4 JNK protein expression was not correlated with gender,age, differentiation(P>0.05).Conclusion:1 JNK mRNA and protein expression is higher in esophageal carcinoma than in esophageal mucosa, which inhibits that JNK plays an important role in the carcinogenesis of esophageal carcinoma.2 JNK protein mRNA and protein expression in esophageal squamous cell esophageal mucosa was significantly higher than those with lymph node metastasis and clinical stage, JNK gene play a role in esophageal cancer and in promoting development.3 JNK may be a clinical diagnosis of esophageal squamous cell carcinoma.
