Objective To evaluate the genotypic distribution of MCP-1-2518A/G single nucleotide polymorphism in the south Han population of Jiang su province and the ethnic differences of this polymorphism. To study the relationship between polymorphism and coronary heart disease and explore the MCP-1 gene polymorphism with degree of the coronary artery stenosis.Methods The single nucleotide polymorphism was detected by using single-labeled probe technique. MCP-1 gene -2518A/G polymorphisms were analyzed in 302 healthy individuals and 297 patients suffered from coronary heart disease confirmed by coronary angiography. The genotype and the allele frequency of MCP-1 was calculated in the different ethnic or the region of the world. The susceptivity gene polymorphism with coronary heart disease was explored and the genotype and the allele frequency of MCP-1 was calculated in the different degree of coronary artery stenosis.Results 1. MCP-1-2518A/G polymorphisms was found in Han people of China. The distribution of genotype of MCP-1-2518 A/G is as follows: 13.6%(41/302),46.7%(141/302)和39.7%(120/302); The genetic frequency of A and G alleles of MCP-1-2518A/G are 36.9%(223/604),63. 1%(381/604), respectively. The distribution of genotype and the allele frequency of MCP-1 is significant different from the people of Slovak Tunisian,Irish and South Africa(P<0.001), but no differences was found between the people in China and in Korea and in Mexican (P>0.05). 2. There were no statistical differences in three genotypes and the allele between coronary heart disease group and normal control group, genotype frequency shows significant association of AG genotype with progressive degrees of coronary artery stenosis (P=0.035), AG carriers had 2.15 times risk for increased severity of coronary artery stenosis compared with AA carrers (P=0.029, 95%CI: 1.10~4.22).Conclusion 1. The ethnic diferences of the south Han population of Jiang su province is significant different from the people of Slovak,Tunisian,Irish and South Africa in the distribution of genotype and the allele frequency; and no differences was detected among the population of Han,Korea and Mexican. 2. AG genotype of MCP-1 is involved in processes leading to enhanced stenosis in atherosclerotic coronary arteries and AA genotype appears to be protective in the Han population of China.
