The Therapeutic Effect Of Two Rattan Mixture On Treating Ankylosing Spondylitis And Changes In Rats Intervention Study Of The Mechanism Of Cartilage

The therapeutic effect of ankylosing spondylitis treated by Chinese medicine is sure.And the toxic action is little,however,up to now,the feature,superiority and mechanism of action of ankylosing spondylitis treated by Chinese drugs preparation is still lack of scientific evaluation.So we carry out this study to offer scientific evidence of the therapeutic effect of Two rattan mixture on Treating Ankylosing SpondylitisObjecive:Evaluate the therapeutic effect of Two rattan mixture objectively which is used to treat ankylosing spondylitis and on its mechanism.Method:1.choose eighty examples which accord with standardment of AS, divide them into two groups randomly,therapy group treated by Two rattan mixture,control group treated by Sulfasalazine.Treat two groups for six months,monitoring blood routine,the function of liver and kidney during treatment,use ASAS20 assessment of ankylosing spondylitis,to observe activity index numbe of pathogenetic condition,total evaluation of patients, total index number of arthrosis swell,physical function,the improval condition of the range of motion of skeleton and muscular system.2.The use of 46 Wistar rats,as a AA model rats were randomly divided into 5 groups: 2 rattan mixture group,indomethacin group,enteric-coated tablets of sulfasalazine group,saline group and normal group.14-day oral treatment; take the side of AA in rats before and after the toe joints,fixed,sliced, HE staining of neutrophils,lymphocytes,plasma cell infiltration,synovial hyperplasia,pannus formation,cartilage and bone destruction evaluated.AA rats before and after taking the side of toe joints,fixed,sliced, immunohistochemistry semi-quantitative detection of TNF-α,MMP-3 and TIMP protein;isolated rat chondrocytes,ⅠL-1βstimulation,the group joined the drug-containing serum,detection of typeⅡcollagen and GAG content of cartilage.Results:1.the satisfaction rate of Two rattan mixture group is 82.5%, Sulfasalazine group is 55%.Compare the therapeutic effect of two groups, P＜0.05.every indicatrion has statistics difference after six mouths' treatment of Two rattan mixture which include body function,activity index number of pathogenetic condition,total swelling arthrosis,total selfassessment, rachialgia,rachiodynia rhachitis,activity of thoracic skeleton, Schober,distance between fingers and ground,left flection of antisternum, right flection of antisternum,antisternum hypsokinesis,distance between head and wall,ESR,C reactive protein;every indicatrion has statistics difference after the treatment of Sulfasalazine except the indicatrion of distance between head and wall.six months later,Compare the therapeutic effect of two groups,indicatrions of total self-assessment and antisternum hypsokinesis have statistics difference,others have no statistics difference.2.The effects of joint damage,the mixture rattan sulfasalazine group and the difference between groups,and GroupⅡdamage rattan mixture sulfasalazine group than the light;Second rattan mixture indomethacin group and group differences;on the expression of TNF-α,two rattan mixture sulfasalazine group and the difference between groups,and the mixture group than in the second rattan sulfasalazine group than weak;Second rattan group and anti-inflammatory agent There were no differences in pain;on the expression of MMP-3,two rattan mixture sulfasalazine group and poor group Different,and two rattan mixture sulfasalazine group than in the weak group; Second rattan indomethacin group and the mixture was no difference.On the expression of TIMP3,two rattan mixture indomethacin group and no significant difference between groups;with sulfasalazine group differences,and the expression of sulfasalazine group than strong.On typeⅡcollagen and GAG content of cartilage,the mixture rattan indomethacin group and group differences;Second rattan mixture with sulfasalazine tablets are different, and two high rattan mixture to sulfasalazine.Conclusion:Two rattan mixture has the certain therapeutic effect of theating AS.And its mechanism of anti-inflammatory and cartilage-related protection.
