The Impact Of Herbal Medicine Of Nourishing Yin Clearing Heat To The Patients Of SLE With Internal Heat Due To Yin Deficiency's Peripheral Blood Mononuclear Cells Apoptosis And Secretion Of IL-2, IL-10

Objective:Systemic lupus erythematosus(systemic lupus erythematosus,SLE) is a serious human health hazards of a typical autoimmune disease,whose etiology, pathogenesis have not yet been fully clarified.It's clinical manifestations is complex and treatment is difficult.Chinese medicine have their own advantages and distinct characteristics on the treatment of SLE.It has multi-site,multi-link and multi-target multi-features as well a significant effect for the treatment of systemic lupus erythematosus which has a characteristic of overall genetic regulation,complex pathogenesis and disease prevention and of is of great significance.We have chosen an effective clinical treatment of systemic lupus erythematosus herbal medicine of nourshing yin clearing heat and carried out the study on SLE patients,and investigate infection of cytokines which was secrete by peripheral blood mononuclear cell.The study can explore the field of cytokine regulation also we can stduy target mechanism that herbal medicine of nourshing yin clearing heat in the role of treat systemic lupus erythematosus.It also can help us deepen understand the mechanism of Chinese medicine in the treatment of systemic lupus erythematosus,and also can help to find effective treatment for systemic lupus erythematosus in the methods of associate with traditional Chinese and western medicine.Methods:1.Containing serum preparation:Preparation Ziyin serum containing Qingre side to side Ziyin Qingre adult dose converted into irrigation Ziyin Qingre gastric side of the rats were administered three days,at the end of the last dose that is administered after the 1h,heart blood,respectively. Stand-membrane filter.2.SLE patients with peripheral blood mononuclear cells(PBMC) isolation and incubation:sterile taken heat deficiency in patients with SLE-type fasting venous blood,after dilution,wind and percussion,centrifugal separation steps, such as PBMC,tissue culture plate by adding 24-plate,under 5%CO2,37℃culture incubator.3.PBMC culture and interfere with detection of apoptosis MTT,flow cytometry of PBMC secreting cytokines.4.The establishment of hormone group,serum-free group,plus hormone medicine group medicine group interventions were observed in different heat of deficiency type of systemic lupus erythematosus patients with peripheral blood mononuclear cells secreted cytokines.Results:1.Apoptosis:hormone plus herbal group,estrogen group of SLE patients can be significantly accelerated in vitro apoptosis of PBMC,suggesting that patients with SLE whose peripheral blood mononuclear cells is very active,beacause SLE patients produce a large number of sef-reactive single-core.Hormone and chinese berbal may be actived by inducing their apoptosis of mononuclear cells respond to and reduce their response to the number of mononuclear cells play a role in treatment.2.The condition of IL-2 secretion on different intervention to the patients with systemic lupus erythematosus.The hormone group,horemone pulse herbal group and blank group were significantly different.Chinese medicine group compare to the blank group was increased but no significant difference. Medicine herbal group and hormones group have a role of activative secretion of Thl-type cytokines cell on SLE patients.The results of this study showed that compared to the secretion of IL-10 herbal medicine of Nourishing yin clearing heat combination with hormone was significantly lower than the blank group.The reduction between herbal medicine of Nourishing yin clearing heat group and the blank group have statistical significance.3.Drugs(medicine,hormones) cause cytokine IL-10 descend and IL-2 is characterized by elevated,IL-10 may inhibit the reduction of the excessive proliferation of B cells in order to reduced autoantibody production. Conclusion:1.On the perspective of cytokine,herbal medicine of Nourishing yin clearing heat has it's own treatment on the type of internal heat due to yin deficiency patients with systemic lupus erythematosus,and the mecanism of therapy may be similarities compare to the hormone's.2.Nourishing yin clearing heat regulate a single cytokine,ulteriorly regulate the overall cytokine network,and it accord with the overall concept of philosophy in the Chinese medicine.At the same time that the mechanism of therapy is complex.3.the study may provide us with new idea on systemic lupus erythematosus prevention and treatment of morbidity.
