Objective To investigate the effect of L– Arg, the substrate of nitric oxide, and L– NAME, the synthase inhibitors of nitric oxide, on P-selectin and inflammatory lesions at the early stage of rats cerebral ischemia-reperfusion.Methods Establish rats middle cerebral artery occasion/-reperfusion (MCAO/R) model, at the same time, make the establishment of sham operation group. Qulified MCAO/R models of rats were randomly divided into three groups as follows: MCAO/R + L - Arg group, MCAO/R + L– NAME group and MCAO/R +saline group, and injected the same volume of L-Arg, L-NAME and saline to the corresponding group at the early stage of rats cerebral ischemia. Detect the content of NO , P-selectin and the activity of MPO at the time point of 4h,8h and 12h of reperfusion, at the same time, and observe the volume of cerebral infarction at 12h of reperfusion using TTC staining.Results 1. find no P-selectin and only detect small amounts of NO and lower MPO activity in the sham operation group.2. find P-selectin at all observed time points and the content of NO was higher than the sham operation group, and the result was significantly different by statistical analysis(P<0.05). Meanwhile, the activity of MPO has the same result at 8h and 12h(8h P<0.05, 12h P<0.01)but the comparison of 4h has no statistic significance(P>0.05).3. The content of NO in MCAO/R +L-Arg gropu was higher than the MCAO/R +saline group at all time point, and the result was significantly different by statistical analysis(4h,8h P<0.05,12h P<0.01). On the contrary , the expression of P-selectin decreased at 8h and 12h between them (8h P<0.05,12h P<0.01),the MPO activity has the same result at all observed time points ( 4h P<0.05,8h,12h P<0.01).simultaneously, the volume of cerebral infarction at 12h of reperfusion by TTC staining was smaller(P<0.01).4. The content of NO in MCAO/R +L-NAME gropu was lower than the MCAO/R +saline group at 8h and 12h(8h P<0.05,12h P<0.01), oppositely , the expression of P-selectin increased at 12h between them ( P<0.05),the MPO activity has the same result at 8h and 12h (8h P<0.05,12h P<0.01).so the volume of cerebral infarction at 12h of reperfusion by TTC staining was bigger(P<0.01).Conclusion 1. P-selectin appears in early inflammatory lesions of cerebral ischemia reperfusion.2. the using of L-Arg and L-NAME can change the amount of NO in the cerebral tissue and then influence P-selectin and the inflammatory processing. So the inflammatory injury of ischemia reperfusion can be alleviated obviously by the supplement of L-Arg.
