Objective To investigate the effect of GSH on ICAM-1 and inflammatory lesions at rats cerebral ischemia-reperfusion.Methods 126 healthy male rats were randomly divided into two groups as follows:sham operated group, MCAO/R+saline group(IR) and MCAO/R+GSH group(GI). MCAO/R +saline group(IR) and MCAO/R+GSH group(GI) were rats middle cerebral rtery occasion/-reperfusion (MCAO/R) models, established by middle cerebral arterie occlusion performed with thread inserted for two hours,then releasing them.The rats of MCAO/R+GSH group were injected GSH into peritoneal at rats cerebral ischemia and 8 hours later;the same volume of saline to the MCAO/R+saline group at same time.Detect the content of ICAM-1 and the activity of MPO at the time point of 12h,24h and 72h of reperfusion, at the same time, and observe the volume of cerebral infarction at 12h of reperfusion using TTC staining.Results 1.Find noICAM-1 and lower MPO activity in the sham operation group.2.Find ICAM-1 in IR group at all observed time points was higher than the sham operation group, and the result was significantly different by statistical analysis(P<0.05).Meanwhile, the activity of MPO has the same result at 12h,24h,72h(12h,24h P<0.05,72h P<0.01).3.The expression of ICAM-1 in GI group was higher than the sham operation group, and the result was significantly different by statistical analysis(P<0.05).Meanwhile, the activity of MPO has the same result at 12h,24h,72h(12h,24h P<0.05,72h P<0.01).The expression of ICAM-1 in GI group was decreased than the IR group at 12h,24h,72h (12h,72h P<0.01,24h P<0.05),the MPO activity has the same result at all observed time points (12h P<0.05,24h,72h P<0.01).simultaneously, the volume of cerebral infarction at 12h of reperfusion by TTC staining was smaller (P<.05).Conclusion 1.ICAM-1 appears in inflammatory lesions of cerebral ischemia reperfusion.2. The result suggest that the using of GSH can reduce expression ICAM-1 and influence the inflammatory processing and restrain the infiltration of neutrophil so as to relieve inflammation after focal cerebral ischemia reperfusion in rats, and plan neuroprotective effect against middle cerebral artery occulusion.
