Objective:Through measuring the content of serum amylase,TNFα,IL-10,SOD and MDA in rats with acute pancreatitis,this thesis aims to investigate the regulatory effect ofω-3PUFAs on acute pancreatitis in rats and discuss its mechanism as well as the existing of lipid peroxidation.Methods:60 rats were randomly divided into 2 groups:sham operated group(SHG,n=15) and acute pancreatitis(AP) group with different treatment(n=45).AP model was established by means of the method on retrograde injection of sodium taurocholate into cholangiopancreatic duct.Fish oil fat emulsion(FOG,n=15 ),standard oil fat emulsion(SOG,n=15) and N.S(NSG, n=15) treatments are give through intravenous respectively.The last step is checking the levels of serum amylase(AMS),tumor necrosis factor-α(TNF-α),interleukin-10(IL-10),superoxide dismutase(SOD),and Malondialdehyde(MDA) in 6h,12h and 24h respectively along with the pathological score of pancreatic tissue.Results:1.The levels of AMS,TNF-α,IL-10 and MDA in acute pancreatitis group increased obviously(p<0.01) while the SOD level decreased(P<0.01).Compared with the sham operation group(SHG),the pathological scores of pancreatic tissue in acute pancreatitis group have changed significantly.2.With the application ofω-3 fish oil fat emulsion,though the AMS in FOG group have no obvious change,the levels of TNF,IL-10 have decreased obviously(p<0.05) and il-10/TNF-αhave increased(p<0.01).This means that the decrease of the body inflammatory response and the increase of anti-inflammatory mechanism.Through the comparison of FOG and NSG, both SOD and MDA have significant differences which show the enhancement of the lipid peroxidation.3.Compared with SOG,the AMS in FOG has no significant difference(p>0.05).TNF-α,IL-10,SOD have decreased obviously(p<0.05) while MDA have increased obviously(p<0.01),the value of IL-10/TNF-αhas increased(p<0.05) and there is no statistic difference with SHG(P>0.05).Conclusion:ω-3 fish oil fat emulsion can inhibit the inflammatory response of AP and enhance the lipid peroxidation with decreased damage to pancreatic tissue.Therefore,ω-3 fish oil fat emulsion treatment is safe and effective.
