Objective To investigate the effects of Remifentanil preconditioning on the expression of TNF-αand ICAM-1 during myocardial ischemia-reperfusion injury in dogs.Try to evaluate the mechanism of the myocardial ischemia-reperfusion injury in dogs and the protect mechanism of Remifentanil preconditioning.Methods 24 healthy hybrid canines were randomly divided into 4 groups: Sham group (nly separation coronary artery left anterior descending, threading and not ligated), ischemic-reperfusion group (saline), Remifentanil 2μg﹒ kg-1﹒ min-1 treatment group; Remifentanil2μg﹒kg-1﹒min-1 treatment group;. Coronary artery ligation of the left anterior descending branch, the 1/3 place to establish the dog myocardial ischemia-reperfusion model. According to time,every group was measured the contents at four time points:before blocked, 30min after ischemis, 1h,2h after reperfusion,anticoagulation 2ml artery extraction, 3000 / minute, centrifugal 15 minutes on the tube placed EP enzyme-linked immunosorbent method of quantitative detection requirements, ICAM-1,TNF -α. HE dyeing optical lens observation myocardial cell structure changesResults Compared with sham group,ischemia-reperfusion group increased the expression level of ICAM -1 and TNF-αin cardiac tissue(p<0.05).Compared with ischemia-reperfusion group, RPC treatment group(2μg﹒kg-1﹒min-1) decreased significantly the expression level of ICAM -1 and TNF-α?HE microscopy confirmed that the pathological changes of cardiac tissue were more slight in RPC treatment(2μg﹒kg-1﹒min-1) than in ischemia-reperfusion group.Conclusion These data suggest that myocardial ischemia—reperfusion can result in intensive activation of ICAM -1 and TNF-α.Early treatment with Remifentanil might be benefit for attenuation of myocardial injury induced by ischemia—reperfusion.And the protective function of RPC treatment group(2μg﹒kg-1﹒min-1) was more reliable。...
