Objective To study the effects of different doses of propofol on the production of Caspase-3,bax and bcl-2 following myocardial ischemia-reperfusion injury of rats, and to investigate its mechanism .Methods One hundred twenty rats were divided into four groups:â‘ Sham groupâ‘¡Ischemia-Reperfusion(I/R) groupâ‘¢Low dose groupâ‘£High dose group .The myocardial samples were studied at 30 min after ischemia and 30,60,120,240 min reperfusion after ischemia.The exprement deployed immunohistochemistry(IHC) in order to detect qualitalively and semiquantitatively the express of Caspase-3, bax and bcl-2.Results In IR group ,LD group and HD group ,after ischemia for 30 min ,the level of Caspase-3 was increased and reached a peak at 60 min after reperfusion, and kept the high level . The rate of bcl-2 and bax were increased after ischemia for 30 min and reached a peak at 60 min after reperfusion, and maintained the high level. In LD group and HD group, propofol could inhibit the activation of Caspase-3 and expression of bax and increase the expression of bcl-2, by which it protected myocardial function . But low dose group was better than high dose group.Conclusion Propofol could inhibit I/R- indused increase in Caspase-3 and bax expression and increase the expression of bcl-2 .And low dose group was better than high dose group.
