Objective: To study on and determination ingredients of photosynthetic bacteria and photosynthetic bacteria preparation, and set up high-fat rats model which in order to screen preliminarily in pharmacodynamics to decrease blood lipids of photosynthetic bacteria preparation.Methods:Experiment 1:①Study on and determination the content of the crude fat, crude fiber, ash, soluble sugar, crude protein, chlorophyll and carotenoid in the photosynthetic bacteria cell refering to literature and national standards;②study on and determination the content of coenzyme Q10 in the photosynthetic bacteria culture medium refering to literature.Experiment 2: To establish a content determination method of free anthraquinone, combinated anthraquinone, total triterpenoid and coenzyme Q10 in the compound preparation transformed by photosynthetic bacteria by UV-Vis spectrophotometry and HPLC.Experiment 3: The rats were fed the high fat diet directly and administrated various photosynthetic bacteria preparation by gastric gavage as the experimental object which in order to study and compare preliminarily in pharmacodynamics to decrease blood lipids of photosynthetic bacteria and photosynthetic bacteria preparation while in order to simplify the prescription of the compound preparation transformed by photosynthetic bacteria.Results:Experiment 1:①The ingredients of the photosynthetic bacteria cell included 1.18% crude fat, 1.79% crude fiber, 2.35% ash, 0.391% soluble sugar, 57.99% crude protein, 3.52mg/g chlorophyll and 1.373μmol/g carotenoid;②the ingredients of the photosynthetic bacteria culture medium included 99.7μg/ml coenzyme Q10.Experiment 2: The ingredients of the compound preparation transformed by photosynthetic bacteria included 5.77μg/ml free anthraquinone, 5.70μg/ml combinated anthraquinone, 9.45μg/ml total riterpenoid and 17.29μg/ml coenzyme Q10.Experiment 3: The level of blood lipids can be decreased significantly by the mixed photosynthetic bacteria culture medium preparation and Danshen agents transformed by Rhodobacter sphaeroides which had better activity of antioxidant and can improve significantly the hemorrheologic characteristics in high-fat rats model. Conclusion:①In the experiment, the ingredients of the photosynthetic bacteria cell and the photosynthetic bacteria culture medium were determined respectively and the content determination method of free anthraquinone, combinated anthraquinone, total triterpenoid and coenzyme Q10 in the compound preparation transformed by photosynthetic bacteria were established in order to control and evaluate the quality of the photosynthetic bacteria and the compound preparation transformed by photosynthetic bacteria;②a unique preparation technology which combined fermentation technology with the Chinese medicine pharmaceutical technology was used to prepare the photosynthetic bacteria preparation which can decrease significantly the level of blood lipids, and it also provides a new idea and method for the research and development of new drugs.
