Objective:Angiogenesis occurs to differrent degree in the process of malignant tumors' growth infiltration and metastasis.Basic fibroblast growth factor(bFGF) and matrix metalloproteinases (MMPs) are two kinds of important factors promoting angiogenesis,which can be secreted by leukemic cells and make the level of bFGF and MMPs high in peripherial blood.By detecting the serum concentrations of bFGF and matrix metalloproteinase-2(MMP-2) in patients with chronic myeloid leukemia(CML),we can study their roles in the diagnosis,progression,and prognosis of CML,and provide theoretic evidence of clinic anti-angiogenesis therapy.Method:Collect 51 serum samples of 42 CML patients and detect the concentrations of bFGF and MMP-2 with enzyme-linked immunoabsorbent assay(ELISA).Compare the serum levels ofbFGF and MMP-2with that in normal control and my eloproliferative disease(MPD) control.The differences of bFGF and MMP-2 levels in different phases of CML patients,the relationship between bFGF level and MMP-2 level;their relationship with gender,age,peripheral blood routine,LDH and the size of spleen. Besides,9.patients in chronic phase were followed for 6 months since the beginning of their disease.Results:The bFGF level in 42 CML patients was 56.403±16.655 ng/L,significantly higher than that in normal control group(32.616±5.654 ng/L)(P<0.001),and had no difference with MPD control group(60.784±16.121ng/L)(P>0.05).The bFGF level in accelerated phase(68.181±2.725ng/L) and blast phase(72.121±9.066ng/L) were much higher than that in chronic phase(50.773±16.056ng/L) (P<0.05),which was higher than normal control group(P<0.001).The MMP-2 level in 42 CML patients was 5190.603±1189.591 ng/L,significantly higher than that in normal control group(2944.134±525.060 ng/L)(P<0.001)and MPD control group(4376.706±1249.011ng/L)(P<0.05).The MMP-2 level in accelerated phase(6093.532±331.207ng/L) and blast phase(6453.878±863.032ng/L) were much higher than that in chronic phase(4745.351±1049.096ng/L)(P<0.01),which was higher than normal control group(P<0.001).There was no difference between MMP-2 level in chronic prase and MPD control group(P>0.05),but in accelerated phase and blast phase the MMP-2 levels were much higher than MPD control group(P<0.001).The bFGF and MMP-2 levels were related to peripheral blood routine,LDH and the size of spleen.There is positive correlation between bFGF level and MMP-2 level in CML patients.Conclusions:The bFGF and MMP-2 levels in CML patients are significantly increased,and the serum levels ofbFGF and MMP-2 in accelarated phase and blast phase are significantly higher than in chronic phase.The bFGF and MMP-2 levels in CML patients are related to the peripheral blood routine,LDH and the size of spleen.To detect the bFGF and MMP-2 concentrations in CML patients is conducive to the diagnosis,assessing,prognosis and directing therapy.
