Effect Of Kang Chan Ning Fang On Peroxidation And DA Neuron Parkinson's Disease Rat Model

Objective: To research the traditional Chinese drug kang chan ning fang to parkinson's disease rat's nerve behaviors ameliorative functions and discuss its functionary mechanism.Methods: all rats keeps a week and then divides eight rats be used as only false surgical operation group, the right side substantia injects 0.9%NaCL; The rest rats builds the models, accordinging to the literature of the rat stereoscopic fixed position diagram, confirm two coordinates of right side substantia, then inject 6- OHDA, two days after makes behavior examines, and records the revolving start time and ending time and the revolving numbers. The successful rat model be divided into three groups with random .Each group have eight rats. We induce the rats with the APO after giving the medicine two weeks, and test the rats revolving the behavior, and record to revolving to start time,ending time and revolving the numbers. Next day open the rat's thorax after proceeding the anesthesia, exposing the heart well, using the syringe pinhead stabs into the left ventricle, taking not anticoagulant whole blood three milliliters, after placing twenty minutes, with the low speed centrifuge blood at 3000 r/ min, leave first the 15 mins, then take the serum, put into the -37℃refrigerator, respectively measure the SOD,MDA and NO in serum with the colorimetry; Taking the perfusion makes the internal fixation, being to see the rat appear the limbs extremities tic,hard, express the perfusion completes, and then cut down the rat head , take brain, soaked in 4% formaldehyde, fix 24 hours in 4℃refrigerator , then wash with the PBS , dehydrates with the gradient alcohol, hyalinize with the xylene, immersing the wax, embed it with paraffin. Paraffin is continuously cut into slices,slices are directly sticked on the carrier slice of the gelatin peridium, select four slices every piece tissue , make all slices to be taked off the wax, then use the PBS to douche, and make them fixed, then use the method of SABC to make the TH immunity tissue chemistry dyes,under the 200 times discrepancy microscopes, add up the number of the male nerve cell in immune response of the total amount right side substantia in difference and calculate its average.Results: comparing with before treating, the traditional Chinese drug group and western medicine group's average revolving speed and keeping on time are all descend and have the statistics meaning. These indicate the traditional Chinese drug group and western medicine group all made the PD rats's clinical symptoms be ameliorated. Average revolving speed and keeping on time of the traditional Chinese drug group are reduced than western medicine group, and have the statistics meaning. These explain traditional Chinese drug group is better than western medicine group in the aspect of PD rat's clinical symptoms ameliorated. Model group and western medicine group are obviously descendent than false surgical group in SOD ( P<0.01), traditional Chinese drug group and false surgical group are indistinctive in SOD( P>0.05).The model group are obviously elevatory in MDA than false surgical group,traditional Chinese drug group and western medicine group ( P<0.05), but the traditional Chinese drug group and the western medicine group are indistinctive in MDA than the false surgical group( P>0.05).The model group are obviously elevatory in NO than the false surgical group,the traditional Chinese drug group and the western medicine group ( P<0.05), but the traditional Chinese drug group and the western medicine group are indistinctive in NO than the false surgical group( P>0.05).The TH cells of the false surgical group,the traditional Chinese drug group and the western medicine group are more than the model group( P<0.01),The TH cells of the traditional Chinese drug group are more than the western medicine group in the right side of the rat's substantia( P<0.05), The TH cells of the false surgical group are more than western medicine group in the right side of the rat's substantia( P<0.01), The TH cells of the traditional Chinese drug group and the false surgical group are indistinctive in the right side of the rat's substantia (P>0.05).Conclusion: Kang chan ning fang and madopar all can obviously ameliorate the revolving behavior of PD rat, but Kang chan ning fang is better than madopar in the aspect of improving parts symptoms.Its treatment mechanism consists in:One is to improve the holistic Hyperoxidation;Two is to reduce the toxin of NO;Three may be to protect DA nerve cell that can be not damaged and correspondingly increase its quantity. Kang chan ning fang is different from Madopar in the aspects of improving the tissue's Hyperoxidation and reducing the toxin of NO and correspondingly increase DA nerve cell quantity.
