ObjectivesTo investigate the effects of low,medium and high dose of Tiaogan Jiedu Recipe on the learning and memory impairment caused by the onset of chronic epilepsy induced by pentylenetetrazol in SD rats and the effect on the expression of butyryl hydroxylase(TH).MethodsSixty SD rats were randomly divided into 10 blank control groups.The other50 intraperitoneal injections of pentylenetetrazol(PTZ)replicated chronic epilepsy model for 28 consecutive days.The blank control group(group A)was intraperitoneally injected with the same amount of normal saline.Subsequently,50 model rats were randomly divided into model group(group B),sodium valproate western medicine control group(group C),low dose group of Tiaogan Jiedu Repice group(D group),midde dose group of Tiaogan Jiedu Repice(E group),high dose group of Tiaogan Jiedu Repice(F group).The blank control group and the model group were intragastrically administered with normal saline.The sodium valproate group was administered with 94.5 mg/kg.d,The low,middle and high dose of traditional Chinese medicine were administered with Tiaogan Jiedu Repice(5.42,10.84,21.68 g/kg~?d),sodium valproate for 28 consecutive days.Observed the seizures of the young rats,the learning and memory scores,and the expression level of TH in the substantia nigra(SN)and ventral tegmental area(VTA).Results1 Young rats with seizure latencyAfter treatment,there was no seizure in the blank control group;the seizure latency of the model group,sodium valproate group,low dose,middle dose,and high dose group of Tiaogan Jiedu Repice is(187.22±19.07),(463.00±42.67),(206.67±31.50),(357.00±44.26),(447.00±36.91);compared with the model group,the seizure latency of the sodium valproate group,the middle dose,high dose group of Tiaogan Jiedu Repice was prolonged,and the difference was statistically significant(P<0.01).compared with sodium valproate group,the seizure latency of the low dose and middle dose group of Tiaogan Jiedu Repice was significantly lower,and the difference was statistically significant(P<0.01).the high dose group of Tiaogan Jiedu Repice was similar,,and the difference was no significant(P>0.05).the seizure latency of the Tiaogan Jiedu Repice low dose and middle dose group was significantly reduced than that of Tiaogan Jiedu Repice high dose group(P<0.01)。2 market test results1)The central residence time of the marketThe central area residence time of the blank control group,model group,sodium valproate group,low dose,middle dose and high dose of Tiaogan Jiedu Recipe was(82.67±9.02),(27.90±7.36),(62.61±7.17),(47.53±8.13),(64.60±9.26),(78.24±7.07);the sodium valproate,the middle dose and high dose of Tiaogan Jiedu Recipe all can significantly prolong the central area residence time of young rats;and The central area residence time ofTiaogan Jiedu Recipe low dose and middle dose goup was significantly lower than that of Tiaogan Jiedu Recipe high dose group(P<0.01).The central area residence time of the blank control group and Tiaogan Jiedu Recipe high dose group was no significantly differencere(P>0.05).2)The total distance of the market The total distance of the market of the blank control group,model group,the sodium valproate group,the low dose,middle dose and high dose of Tiaogan Huazhuo Jiedu Recipe was(22.41±3.25),(6.72±1.74),(13.56±3.89),(11.45±2.35),(14.99±2.62),(19.99±4.64);the sodium valproate,low dose,middle dose,high dose of Tiaogan Jiedu Recipe can significantly increase the total distance of the youngrats.The total distance of the of Tiaogan Jiedu Recipe low dose and the middle dose group was significantly lower than that of the Tiaogan Jiedu Recipe high dose group(P<0.01).The was no significant differencere between the blank control group and Tiaogan Huazhuo Jiedu Recipe high dose group of the total distance(P>0.05).3 water maze test results1)The navigation resultsThe water maze escape latency period of the blank control group,the model group,the sodium valproate group,the low dose,middle dose,high dose group of Tiaogan Huazhuo Recipe,The first day was(33.90±4.22),(50.48±5.24),(44.55±5.70),(47.07±5.38),(40.20±3.95),(35.79±3.74),and the next day was(24.16±3.54).(41.13±5.53),(39.51±5.54),(35.87±3.56),(31.52±4.62),(24.64±4.05),and the third day were(12.85±4.30),(35.80±7.80),(29.03).±6.29),(29.69±4.00),(19.14±5.38),(13.07±3.27),and the fourth day were(8.90±3.44),(23.07±6.45),(17.53±6.32),(17.11±5.09).(10.95±4.25),(10.47±6.92),and the fifth day were(5.95±2.06),(14.28±3.67),(11.69±3.27),(11.73±2.16),(6.99±1.83),(5.90).±1.43);the sodium valproate,low dose,middle dose,and high dose of Tiaogan Jiedu Recipe can reduce the varying degrees of escape latency of young rats,the middle dose and high dose of Tiaogan Huazhuo Jiedu Recipe can significantly reduce the escape latency of young rats(P<0.01),and the escape latency of the Tiaogan Huazhuo Jiedu Jiedu Recipe middle dose and high dose group was significantly shorter than that of Tiaogan Jiedu Recipe low dose group(P<0.01).2)Results of space explorationThe number of times of the blank control group,the model group,the sodium valproate group,the low dose,middle dose high dose group of Tiaogan Huazhudu Recipe is(6.67±1.00),(2.67±1.32),(3.44±1.42),(3.56±1.51),(5.11±1.27),(6.11±1.36).The middle dose and high dose of Tiaogan Huazhuo Jiedu Jiedu Recipe can significantly increase the number of plateau passages in epileptic SD rats(P<0.01),and the number of plateau passages of middle dose and high dose of Tiaogan Jiedu Recipe was higher than that of the low dose of Tiaogan Jiedu Recipe(P<0.05),There was no significantly differencere between Tiaogan Jiedu Recipe middle dose and high dose group(P>0.05),which was less than the number of times the blank control group crossed the platform(P<0.05).There was no significantly differencere between the Tiaogan Huazhuo Recipe high dose group(P>0.05).4 TH expression results1)Substantia nigra The number of TH positive expression in the substantia nigra of the blank control group,the model group,the sodium valproate group,the low dose,middle dose,high dose group of Tiaogan Jiedu Recipe was(38.50±6.75),(12.70±4.11),(35.00±6.99),(17.30±5.61),(27.90±7.92),(34.90±7.78),the sodium valproate,middle dose,and high dose of Tiaogan Huazhuo Jiedu Recipe can significantly increase the number of TH cells in the substantia nigra(P<0.01),The number of TH positive expression in the substantia nigra of the Tiaogan Huazhuo Recipe low dose group and middle dose group was lower than that of the Tiaogan Huazhuo Recipe high dose group(P<0.01,P<0.05).The number of TH positive expression in the substantia nigra of Tiaogan Jiedu Recipe high dose group was no significantly differencere between the black control group and the sodium valproate group(P>0.05).2)The ventral tegmental area The number of TH positive expression in the the ventral tegmental area of the control group,the model group,the sodium valproate group,the low dose,the middle dose,and the high dose group of Tiaogan Jiedu Recipe was(29.40±9.43),(14.40±5.42),(16.00±5.93),(14.80±4.67),(18.80±6.11),(28.60±7.97).high dose of Tiaogan Huazhuo Jiedu Recipe can significantly increase the number of TH positive expression in VTA of young SD rats with epilepsy(P<0.01),The expression of TH positive cells in the VTA of the Tiaogan Huazhuo Jiedu Recipe low dose group and middle dose group was significantly lower than that of the Tiaogan Huazhuo Recipe high dose group(P<0.01).Conclusions1 Tiaogan Jiedu Recipe can significantly prolong the incubation period of seizures in young rats with chronic epilepsy induced by PTZ,and can significantly improve the learning and memory ability and reduce the anxiety level of epileptic young rats.There was a dose effect between the low,middle and high dose of Tiaogan Jiedu Recipe.The high dose of TiaoganHuazhuo Jiedu Recipe improved the onset of epilepsy and improved the learning and memory effect.2 Tiaogan Huazhuo Jiedu Recipe can significantly increase the number of TH positive neurons in the substantia nigra and VTA of PTZ-induced epilepsy model rats.The effect of Tiaogan Jiedu Recipe high dose group was the most obvious.It is suggested that Tiaogan Jiedu Recipe may play a role in promoting dopamine synthesis in the brain of young rats.3 Tiaogan Jiedu Recipe has
