Objective:To observed the effects of BP on SOD,MDA,BCL-2,apoptosis in a number of brain cells after CIRI of brain tissue in mongolian gerbil,on the basis of selection of BPH,BPM,BPL doses of the three treatment groups were compared,preliminary study of its effect is dose-dependent existence。Methods:120 male Mongolian gerbils in 8 week old,in the environment of normal feeding adaptability for a week,were randomly divided into group S group,M group,BPH,BPM,BPL5 groups,24 in each group。At the beginning of the experiment the first day of the S,M group were given,the same amount of saline gavage,Once a day,BPH,BPM,BPL group were given 300mg/kg/d,200mg/kg/d,100mg/kg/d gavage,Gavage once a day in each group,operation model after two weeks。Modeling methods:The use of temporary clipping first gerbil bilateral carotid artery lax approach to create gerbil cerebral ischemia reperfusion injury model;Model concrete steps:M,BPH,BPM,BPL group after bilateral carotid artery occlusion 10min, release artery surgery and Surgical suture,S group only isolated gerbil bilateral carotid artery without clipping bilateral common carotid artery。After the Success of the model continues to repeat,the administration of pre-operative way,After administration, respectively at 24h,72h,120h different time points in each group after opimus perfusion in vivo(4) and non-perfusion in vivo(4),having broken two kinds of death,Take one in four non-perfused gerbils of fresh brain tissue,determination of cerebral infarction through the area of TTC staining,UV Spectrophotometric Determination of SOD,MDA contents。In addition,after taking four of gerbil in the perfused rat brain tissue,Through DAB staining(chemical method) Determination of suppressing apoptosis gene Bcl-2 positive cell count and one-step TUNEL Apoptosis Detection Kit(immunofluorescence assay) to detect nerve cells wither death of a few。 Results:1,The experimental results show that BP can reduce infarct volume after CIRI,Significant role in BPH group compared with the M group(P<0.01);BPL group compared with the M group reduced infarct volume significantly(P<0.05)。2,On the role of SOD:The SOD content in the treatment group of BP higher than the M group,which increased more significantly in BPH,BPM group than in M group(P<0.01), BPL group compared with the M group has also increased(P<0.05), that enhanced BP CIRI Gerbil brain tissue SOD content of free radical oxidation of brain tissue injury。3,On the role of MDA:BPH,BPM group of MDA content compared with the M group decreased significantly(P<0.01),compared with the S group was not significant,BPL group compared with the M group were statistically significant(P<0.05),show that BP can after CIRI Gerbil brain tissue MDA levels returned to pre-operative physiological level。4,BP of CIRI gerbil brain tissue apoptosis inhibitory gene Bcl-2 expression:The experiment showed that BPH,BPM,BPL group able to improve the CIRI Gerbil brain tissue apoptosis inhibitory gene expression,BPH,BPM group Bcl-2 positive cells than the expression of the M group increased significantly (P<0.01),BPL group Bcl-2 positive Cells than the M group expression,increased(P<0.05),but BPL compared with higher dose group With the extension of time CIRI.in brain tissue Bcl-2 positive cells in the trend has slowed down。5,BP on the brain nerVe cell apoptosis(TUNEL stop method): BPH,BPM group apoptosis positive cells than the M group significantly reduced(p<0.01),BPL group apoptosis positive tells than the M group reduced(p<0,05),the experiment also showed that although the BPH,BPM,BPL group in the reduction of brain neurons CIRI Apoptosis obvious difference between the quantity,but With the extension of time after CIRI reduce brain neurons trends are enhanced。Conclusion:BP have apparent therapeutic effect to Gerbil model about after CIRI,to reduce the volume of cerebral infarction,enhance the content of SOD in brain tissue and reduce the content of MDA,which has played a very good role on the antioxidant of free radical damage;BP treatment group able to improve the CIRI gerbil brain tissue Bcl-2 expression and lower brain neuron apoptosis;And BPH,BPM,BPL group to SOD,MDA,Bcl-2,Apoptosis of brain cells has the role of dose-dependent nature。At the design time window of laboratory,the Bowring paste at increased SOD content of antioxidant free radical damage,reduce the lipid peroxidation product MDA have slowed the trend,but with the time after CIRI to promote the expression of Bcl-2,reduce the apoptosis of neurons cells in brain tissue and the volume of cerebral infarction show the trend are increased,this surface inconsistency shows that BP against CIRI means have a variety of ways,have a persistent effect,which consistent with the characteristics the diversity of element in BP and efficacy of Chinese medicine has a slow but long-lasting。As these laid the experimental foundation for research the BP against oxidative damage,apoptosis of nerve cells and other pathogenic mechanisms after CIRI。...
