Object:Under the guidance of Chinese medicine theory, this experiment takes use of modern medical technology research of Astragalus Polysaccharide influnce on rat cerebral ischemia reperfusion brain tissue pathological histology, Caspase-3, Bax, Bcl-2 protein and SOD, MDA, providing a theoretical basis for application of Astragalus Polysaccharides in clinical ischemic encephalopathy.Methods:72 SPF level Wistar rats were randomly divided into 6 groups, sham operation group, model group, positive control group, Astragalus membranaceus high dose group, middle dose group and low dose group,12 rats in each group; With thread embolism method modified copy of cerebral ischemia reperfusion model of rats, the drug group was respectively given corresponding medicine for 15 days. Quality the brain tissue and calculate the cerebral index of ececuted rats, observe the pathological changes of brain tissue in rats of each group by HE staining.The expression level of Caspase-3, Bax, Bcl-2 protein in brain tissues of rats was detected with Immunohistochemical method, SOD, MDA in the content of brain tissue in rats of each group were detected with xanthine oxidase and Thiobarbituric acid (TBA) method.Result:1.Effect of organ indices and Pathological morphology:the organ indices was hjgher in model group than in Sham-operated group (P<0.05), no statistically significant differences among the other groups, nerve cells of rats in the Sham-operated group was not obvious dropsical, Neuronal cytoplasm cells were rich and nucleus are clearly visible. Arrangement of Nerve fiber bundles were consistent and not disorder. A large number of cribriform necrotic can be seen in model group, neurons Large-scale disappeared, interstitial become loose. compared with the model group, the degeneration and necrosis of nerve cell in drug group presented lesser degree, the area of damaged nerve cell is less, no significant formation of infarct area, the arrangement of Nerve fiber bundles can be observed. The degree of lesions in positive control group and Astragalus membranaceus high dose group was less, too.2. Effects of cell apoptosis:①Caspase-3 expressions:Compared with Sham-operated Group, model group increased significantly (P<0.01).compared with control group, Astragalus membranaceus high dose group and middle-dose group expression was significantly reduced (P<0.05).③Bcl-2 expressions:model group increased significantly than the Sham-operated Group (P<0.05). Astragalus membranaceus high dose group and middle-dose group increased significantly than the model group (P<0.05).③ expression of Bax Protein:expression of Bax Protein in model group increased significantly than the Sham-operated group (P<0.05).compared with control group, expression of Bax protein in Astragalus membranaceus high dose group was significantly reduced (P<0.05).④Compared with the Sham-operated group, Bcl-2/Bax ratio in model group was decreased significantly. compared with Astragalus high, middle, low dose groups, the ratio in positive control group was significantly higher. 3. Effects of lipid peroxidation:①expression of SOD:Compared with the Sham-operated group, SOD expression levels in rat brain tissue in model group were significantly reduced (P<0.05).Compared with model group, groups showed significant difference except the Astragalus membranaceus low dose group (P<0.05).②expression of MDA:Compared with the Sham-operated group, MDA levels in rat brain tissue in model group was increased(P<0.05). Compared with control group, MDA levels was decreased in Astragalus membranaceus high dose group (P<0.05).Conclusion:Astragalus membranaceus has a positive protective effect on cerebral ischemia-reperfusion in rats.The mechanism may be to reduce the extent of apoptosis in neuronal, increased the SOD levels of cerebral ischemia-reperfusion injury rats, decrease the MDA content, improve the pathological changes of the brain.The effect in Astragalus polysaccharides higher doses group than middle, low-dose groups, indicating that the effect was dose-related.
