ObjectivesMalignant ovarian tumour is one of three malignant tumour in female reproductive organs,Epithelium ovarian carcinoma is the most common category.Five survival rate of malignant ovarian tumour is lower,Five survival rate of earlier period ovarian carcinoma is over 75%,but Five survival rate of advanced period ovarian carcinoma with killing tumor cells and chemotherapy is 15-20%.Early diagnosis of ovarian carcinoma is very difficult for many reasons,so criticality of prognosis is early diagnosis and early treatment.PAX2 is one of nine PAX genes,and each PAX gene contains a served paired box domain.The PAX2 gene encodes a transcription factor that plays a critical role in the development of the urogenital tract,eyes,ears,and central nervous system and it is important tumorigenesis.Research point out that PAX2 relates to renal carcinoma,carcinoma of urinary bladder,breast cancer and endometrial cancer.It have been reported that PAX2 expresses in papillary serous carcinoma of the ovary,but it is still unclear if there is different expression in normal ovarian tissue,optimum ovarian tumor and commissurotome ovarian tumor.We used the immunohistochemistry technique and reverse transcription polymerase chain reaction(RT-PCR)to detect the expression of PAX2 mRNA and protein,and provided a theory base for gene diagnosis and treatment of ovarian tumor.MethodsSamples were taken in sheng jing hospital of china medical university during January of 2007 to December of 2007,including 3 lepthelial ovarian cancers,11 normal ovarian tissues,10 optimum ovarian tumors and 11 commissurotome ovarian tumors. Fresh samples were obtained from each patient at the time of surgery.The samples were immediately frozen in liquid nitrogen and were stored at-70℃. RT-PCR and immunohistochemistry technique were used to assess the expression of PAX2 mRNA and protein in 31 epthelial ovarian cancers,11 normal ovarian tissues,10 optimum ovarian tumors and 11 commissurotome ovarian tumors.Statistical analysis was performed using SPSS for window 13.0 software.The comparisons of means were carried out by analysis of variance and comparisons of rates were carried out by X2 test and fisher exact probabilities.ResultsThe results showed the exact biological effects of PAX2 in ovarian cancer tissues from RT-PCR and immunohistochemistry technique.There was no statiatically significant relationgship between gene expression at surgery or clinical-pathologiical staging,P>0.05;there was significant difference between G1-G2 and G3,P<0.01;there was significant difference between advanced stage(Ⅲ,Ⅳ)and early stage(Ⅰ,Ⅱ),P <0.05.significant differences were noted between epthelial ovarian cancer,normal ovarian tissue,optimum ovarian tumor and commissurotome ovarian tumor, P<0.01;there was no Significant differences between normal ovarian tissue,optimum ovarian tumor and commissurotome ovarian tumor,P>0.05.Conclusions1.PAX2's expression is higher in epthelial ovarian cancer than those in commissurotome ovarian tumor,optimum ovarian tumor and normal ovarian tissue.PAX2's expression is higher in epthelial ovarian cancer of advanced stage(Ⅲ,Ⅳ) than that in early stage(Ⅰ,Ⅱ)and PAX2's expression is higher in G3 than G1-G2.2.PAX2 maybe play a suppression role in diagnosis and malignancy appraisal of ovarian cancer.
