ObjectiveBy comparing the hepatic blood inflow occlusion without hemihepatic artery control,semi-hepatic vascular occlusion and Pringle treated animals respectively set up the classic animal model of liver regeneration,and analyzing hepatic blood inflow occlusion without hemihepatic artery control on liver regeneration function in rats.Methods120 male SD rats,weighing 200~250g were randomly divided into hepatic blood inflow cclusion without hemihepatic artery control group(WH group),semi-hepatic vascular occlusion group(HH) and Pringle group(PR group),each group using the corresponding methods of hepatic occlusion rats liver for 15 minutes。with bulldog clamp we resect median lobe,left internal lobe and left outer lobe(70%hepatectomy) to induce liver regeneration.Eight rats in each group were killed at 6 hours,12 hours,24 hours,4 days and 7 days after hepatectomy,respectively.The liver to body weight Ratio,Cells count undergoing mitosis in histological section,and at different time point is done in order to assess the liver regeneration.By detecting expression of CyclinD1, serum concentration of IL-6 were measured at each time point,we can value the effect on liver regeneration were measured at each time point.rusult1.Liver to body weight ratio24 hours the liver weight had no significant differences in body weight in each group(P>0.05);at 4 days in WH group liver to body weight greater than in PR group (P<0.05),and wih the HH group were no significant differences(P>0.05);at 7 days in WH groupliver to body weight greater than in PR group(P<0.05),and with the HH group were no significant differences(P>0.05)2.Hepatocyte Mitotic count(Index)At 24 hours after operation hepatocyte mitotic count in WH groupmore than those in PR group(P<0.05),and with HH group hepatocyte Mitotic count is no significant differences(P>0.05);At 4 days after operation hepatocyte mitotic count in WH group more than those in PR group(P<0.05),and with HH group was no significant differences(P>0.05);at 7 days after operation,there were no difference among the groups(P>0.05).3.Expression of CyclinD 1There was no difference in every group at 6 hours after operation(P>0.05). Compared with PR group,the expression of CyclinD1 in HH and WH group was significantly higher at 12 hours and 24 hours after operation(P<0.05).There was no difference between HH and WH group(P>0.05).4.IL-6the level of IL-6 had reached a peak at 6 hours partial hepatectomy in each group, and gradually reduced within 24 hours.Compared with PR group,the level of IL-6 in HH and WH group was significantly higher at 6 hours and 12 hours after operation(P<0.05).There was no difference between HH and WH group.,while the WH group and the HH group were no significant differences(P>0.05).There was no significant difference at 24 hours,but still higher than the normal group(P<0.05).Conclusion1.The hepatic blood inflow occlusion without hemihepatic artery control in the promotion of liver regeneration in rats with semi-hepatic vascular occlusion has the same effect,is superior to Pringle.2.The upregulated IL-6 and high expression of CyclinD1 may be the one of the reasons why Hepatic blood inflow occlusion without hemihepatic artery control method affects the hepatic regeneration of rats.
