Objective:To approach the effects and related mechanism of Poria combined with aerobicexercise on preventing atherosclerosis in rats, and to provide a theoretical basis for thedevelopment of new prevention and cure of atherosclerosis.Methods:60 male 12-week-old Sprague-Dawley rats were randomly and equally divided into the normal group(C), high-cholesterol group (HC), high-cholesterol+Poria group (M), high-cholesterol+normal saline(SC),high-cholesterol+ Poria+ aerobic exercise group (ME) and then were fed for 10 weeks. With the exception of the normal group (C), all rats were a single dose of intraperitoneal injection of vitamin D3(600 000 IU/kg) before with high fat diet.The pathological changes in the aorta were observed using HE and Weigert staining.Serum triglyceride(TG), high density lipoprotein cholesterol (HDL-C)and low density lipoprotein cholesterol (LDL-C) were detected in enzymatic determination.Immunohistochemistry staining were performed to assess the ICAM-1, LFA-1 and TF protein expression,the expression level to be described in optical density value (IOD). All data were analyzed by SPSS16.0.Results:1.The levels of serum TG,LDL-C in ME group were obviously lower than those in HC,SC group(P<0.01)and M group(P<0.05); The levels of serum HDL-C in ME group were obviously higher than those in other group(P<0.05).2. The ICAM-1, TF expression in aortas in ME group was significantly lower than those in HC,SC group(P<0.01)and M group(P<0.05).3. The LFA-1 expression in aortas in ME group was significantly lower than those in HC, SC group(P<0.01),but there were no significant difference between ME group and M group.Conclusions:1.The levels of serum LDL-C and TG in high-fat diet rats can be actively suppressed by the intervention of Poria combined with aerobic exercise,and the level of HDL-C was increased.2.Protein expression of ICAM-1, LFA-1 and TF were significantly inhibited on aortic wall of high-fat diet rats by the intervention of Poria combined with aerobic exercise.3.The development of atherosclerosis caused by high-fat diet in rats can be delayed via by both Poria and Poria combined with aerobic exercise, the effect of joint action was better than single.
