Objective:In our study,we analyze the workers' chromosome aberration and micronucleus who exposed to electronic waste for a long time and with adverse pregnancy(Missed abortion,Fetal anomaly and Stillbirth fetus) combining with cytokinesis-block micronucleus and microelement test in order to estimate the degree of genome damage and explore the damaging effects of electronic pollutants on reproduction function of the local people.Methods:20 couples(study group) were selected from the villages(live radius≤5 kilometers)where there have been massive disposal of electronic wastes and with averse pregnancy.40 villagers(Male 20,Female 20) lived far from the electronic waste processing regions(live radius range 20 to 50km) were chosen as the control group.Chromosomal aberrations and cytokinesis-blocking micronucleus were performed to detect the cytogenetic effect(dicentric and ring chromosome,acentric fragments,minute chromosomes,chromatid break,chromatid gap),chromosomal aberrations rate and micronuclear rate were scored for each subject.DNA damage was detected using single cell gel electrophoresis,the DNA percentage in the comet tail(TDNA%),tail moment(TM),and Olive tail moment(OTM) were recorded to describe DNA damage to lymphocytes.We also test microelement contents in the peripheral blood in order to known the harmful influence of electronic waste. Chi-Square test and T-test were employed to do statistic analysis.Results:The dicentric and ring chromosome rates was 0.175%±0.004,acentric fragments was 0.1%±0.003,minute chromosomes was 0.225%±0.005,chromatid break was 0.15%±0.004,chromatid gap was 0.075%±0.003%and micronuclear rates was 13.63‰of the study group were significantly higher than in the control group (P<0.001).The percentage of DNA in the comet tail was 6.4107%±3.53743,tail moment was 1.5883%±1.36788,and Olive tail moment was 1.9329%±1.03982,all the data which detected by single cell gel electrophoresis showed that there was a significant difference in DNA damage in the exposure group(P<0.001).The micronucleus rate observed in women was significantly lower than those in men.The contents of zinc,lead,iron and magnesium were significantly higher than in the control group(P<0.001).Conclusion:The use of outdated(and unsafe) ways to deal with E-wastes can lead to exposure to a variety of substances which harmful to human health.The components of pollution may be a potential agent of genetic mutation and may enter the human body through the air,drinking water,and food chain to damage human genetic material,resulting in genomic instability,even inducing the outcome of averse pregnancy.These findings need to be considered,and steps should be taken to protect the current population and future generations from the effects of pollution with E-wastes.We also analyze the reason that E-wastes can induce genetic substances damage,the mechanism and consequence.It will be useful to genetic counseling and clinical diagnosis.
