[Objective]:To explore the expression of tumor suppressor PTEN and hypoxia-inducible factor HIF-1αin different grading gliomas and investigate the correlation between them.[Methods]:Forty-two surgical cases of gliomas from 28 males and 14 females in The Third Clinic College of Kunming Medical College during 2006.3~2008.5 were selected.Age ranged from 15 years to 53 years,average 41.2 years.Controlled group of eight cases came from internal decompression in brain contusion.The age ranged from 18 years to 42 years,average 28.3years.The s-p immunohistochemical method was used to measure the expression of PTEN and HIF-1αin 42 cases of gliomas.[Results]:1.①The total positive rate of the expression of PTEN in gliomas and normal brain tissues were 50.0%(21/42) and 100%(8/8).There was statistically significance(P=0.006) between them.②From gliomas of gradeⅠ~ⅡtoⅢ~Ⅳ, PTEN positive expression decreased gradually,the positive rate were73.7%(14/19) and 30.4%(7/23),they were statistically significance(P=0.001) between two groups. 2.①The total positive rate of expression of HIF-1αin gliomas was 64.3%(27/42). However,it was 0.0%(0/8) in normal brain tissue.There was statistically significant (P=0.001) between them.②The positive rate of expression of HIF-1αinⅠ~Ⅱgrades of gliomas was 42.1%(8/19).However,it was 82.6%(19/23) inⅢ~Ⅳgrades of gliomas.There was statistically significant(χ~2=7.435,P=0.006) between two groups.3.There were 7 cases PTEN-positive of HIF-1α-positive group,whereas 20 cases PTEN-negative of HIF-1α-positive group.The expression of PTEN in gliomas is inversely correlated with the expression of HIF-1αin gliomas(r_s=-0.646,χ~2= 17.526, p<0.05).[Conclusions]:1.The experiment intend to measure the expression situation both of PTEN and HIF-1αin humans' brain tissue and analyse the relative correlation of PTEN and HIF-1αabout gliomas in this study.The expression of PTEN in normal brain tissue is as usual,but along with the pathology grade of glioma becoming higher, the positive rate is decreasing obviously.The The expression of HIF-1αin gliomas is highe than in normal brain tissue.Moreover,along with the pathology grade of glioma becoming higher,the positive rate is increasing obviously.To analyse the correlation in expression of PTEN and HIF-1αis a significant negative correlation.These evidence demostrate that PTEN and HIF-1αare much related to the apoptosis and angiognenesis of glioma and the degree of benign,as well as both of them play a very important role in the mechanism of generation and development in glioma.PTEN and HIF-1αmay be to become the promising tumor mark and new target of genic treatment in gliomas.
