Object to investigate the expression of p27kip1and cyclinD1 in the skin of patients with psoriasis vulgaris,their correlation and their significance in the development of psoriatic lesions.Methods 48 outpatients with psoriasis vulgaris(who were diagnosed by clinic and pathology and had typical lesions)were studied,including 30 cases in progressive stage and 18 cases in static stage.30 cases of normal skin were chosen as controls.Immunohistochemistry was used to study the expression of p27kip1(cyclin dependent kinase inhibitor)and cyclinD1. SPSS 11.5 statistical analysis software was carried out for statistical treatment.Results①p27kip1protein expression Positive expression of p27kip1protein was located mainly in nuclear and seldom in cytoplasm.Brown-yellow or brown positive particles were observed.The expression of p27kip1was significantly lower in lesions with psoriasis vulgaris than in the normal skin controls(P<0.01).The level of p27kip1decreased in static lesions in comparison with the normal skin controls(P<0.05).The level of p27kip1significantly decreased in progressive lesions in comparison with the normal skin controls(P<0.01).There was difference in lesional skin between progressive stage and static stage(P<0.01).②cyelinD1 protein expression Positive expression of cyclinD1 protein was located in nuclear. Brown-yellow or brown positive particles were observed.There was significant increase of cyclinD1 in lesions with psoriasis vulgaris in comparison with those of the normal skin controls (P<0.01).The level of cyclinD1 increased in static lesions in comparison with the normal skin controls(P<0.01).The level of cyclinD1 increased significantly in progressive lesions in comparison with the normal skin controls(P<0.01).And the expression of cyclinD1 was higher in lesions of progressive stage than of static stage(P<0.05).③There was negative correlation between p27kip1and cyclinD1 in psoriasis vulgaris(r=-0.302).Conclusion①The down-regulation of p27kip1is likely to relate to overproliferation of keratinocytes in psoriatic lesions.②The expression of cyclinD1 was significantly higher in psoriatic lesions than that in the controls.It was also significantly higher in progressive lesions than that in static lesions.CyclinD1 is a factor in evaluating hyperplasia of keratinocytes in psoriasis vulgaris.③In the progress of psoriasis vulgaris,they regulated each other.
