Purpose:? To detect the relevant between Single nucleotide polymorphism of S100A4 mRNA 98A/T,mRNA 208G/T and the first intron of S100A4 methylation status and their effect on the occurrence and lymph metastasis of the early stage of cervical cancer in Shanxi .Methods: A retrospective case-control study design was applied for the present study. 1. The control group consisted of 178 normal paraffin embedded cervical tissue samples and 186 of peripheral blood samples from healthy women. As the same time, the case group included 182 paraffin embedded cervical tissue samples (135 cases without metastasis and 47 cases with lymph metastasis) and 213 peripheral blood samples (170 cases without metastasis and 43 cases with lymph metastasis) from patients with the early stage of cervical cancer.Both groups of samples were collected from Shanxi province. Using genomic DNA as templates, we employed Tetra-primer amplification refractory mutation system polymerase chain reaction (ARMS-PCR) to analyze the genotype distribution and allele frequency of S100A4 mRNA 98A/T,mRNA 208G/T. Chi-square test ( 2 test) was used for statistical comparison of genotype distribution and allele frequency between the control group and the case group as well as between the group without metastasis and the group with metastasis. Odds ratio (OR) and 95% confidence interval (CI) indicated relative risk. 2.Using methylation-specific polymerase chain reaction (MSP) to detect the first intron of S100A4 methylation status in 21 normal paraffin embedded cervical tissue samples and in case group included 123 paraffin embedded the early cervical tissue samples (63 cases without metastasis and 60 cases with lymph metastasis).Chi-square test ( 2 test) was used for statistical comparison of the first intron of S100A4 methylation status between the control group and the case group as well as between the group with metastasis and the group without metastasis. Results: 1. S100A4 mRNA 98A/T Single nucleotide polymorphism include three genotypes: AA,AT and TT. S100A4 mRNA 208G/T Single nucleotide polymorphism include three genotypes: GG,GT and TT.①In peripheral blood samples, for S100A4 mRNA 98A/T gene polymorphisms including genotype distribution and allele frequency, there is no statistical significance between the control group and the case group (P>0.05),but there is statistical significance between the group without lymph metastasis and the group with metastasis(P<0.05), Compared to A allele, T allele maybe increase the risk of cervical cancer(OR=2.117,95% CI=1.205-3.722);as the same time,S100A4 mRNA 208G/T gene polymorphisms including genotype distribution and allele frequency has no statistical significance between the control group and the case group (P>0.05),also there is no statistical significance between the group without lymph metastasis and the group with metastasis(P>0.05).②For S100A4 mRNA 98A/T,mRNA 208G/T gene polymorphisms including genotype distribution and allele frequency, in tissue samples, there is no statistical significance between the control group and the case group as well as between the group without lymph metastasis and the group with metastasis (P>0.05). 2. Using the MSP method to detect the control group and the case group as well as between the group without lymph metastasis and the group with metastasis, we found that the first intron of S100A4 methylation status has statistical significance (P<0.05) .Conclusion:? 1.S100A4 mRNA 98A/T Single nucleotide polymorphism with the occurrence of cervical cancer had no significant correlation,but in peripheral blood,when the A→T base change of S100A4 mRNA98 may increase lymph metastasis of early cervical cancer.2.there has the relevant between the low methylation tend of S100A4 first intron and the occurrence and lymph metastasis of the early stage of cervical cancer.
