Objective The aim of the present study was to teste long term neuroprotective effect of intraperitoneal application of saturated hydrogen saline in an established neonatal HI model.Methods The model used in this study is based on the Rice-Vannucci model. 2,3,5-triphenyltetrazolium chloride(TTC),HE and Nissl straining were preformed to evaluate the effect of hydrogen saline.(HS).Immunohistochemistry was used to observe the number of TUNELL-positive cells in damaged cortex and hippocampus of the different groups.The amounts of caspase-3 activities were analyzed by Caspase-3/CPP32 Fluorometric Assay Kit.At 5 weeks after HI,Body weight,function test,spontaneous activity tes tand Morris water maze test were conducted for the long-term neurological and neurobehavioral functions;Results Reserch on the effect of hydrogen saline:comparison with the ischemic control group,hydrogen saline intraperitoneally can reduce the cerebral infarction area,increased the survival of nerve cells,reduce the Caspase-3 activity,and significantly increase thelong-term learning and memory in rats.Conclusions peritoneal administration of H2 saline reduces brain injury after neonatal HI possibly by attenuating oxidative stress,leading to reduction of apoptosis and improvement of long term neurological and neurobehavioral functions.
