The thesis studies on the constituents and anti-tumor activity in vitro of Aconitum vaginatum Pritz.. Seventeen compounds are isolated from its ethanol extract. Fifteen compounds'structures of them are identified by spectroscopic analysis (IR,EI-MS,1H-NMR,NOSEY,13C-NMR,DEPT,1H-1H COSY,HSQC,HMBC, et al.). Seven of them are from the alkaloids layer: 2-(acetylamino)-benzoic acid, methyl ester (Av7), 2-[2'-(2''-hydroxy-2''-methyl-propionylamino)-benzoylamino]-benzoic acid methyl ester (Av9), 2-[2'-(2''-hydroxy-2''-methyl- butanylamino)- benzoylamino]-benzoic acid methyl ester (Av12), cammaconine (Av8), atisinium chloride (Av11), vaginatu- nine (Av16), lappaconitine (Av17); and others are non- alkaline substances. Their structures are established respectively to beβ-sitoterol (Av1), daucosterol (Av2), hexadecanoic acid (Av3), hexadecane (Av4), 3, 4-dihydroxybenzaldehyde (Av5), p-hydroxybenzaldehyde (Av6), p-hydroxybenzoic acid (Av13), benzoic acid (Av15). Among them, Av1-Av3, Av7, Av8, Av11 and Av17 are first isolated from this plant; Av4-Av6 are first isolated from the genus Aconitum; Av9, Av12, Av16 are new.To screen active layers of Aconitum vaginatum and active compounds for anti-tumor activity in vitro, MTT reduction test is designed. The experiment observes the samples'inhibition rate to AGS,Hepg2,A549 of cell strains. Total extract and alkaloid layer are detected to have apparente activity. Meanwhile, Av7, Av8, Av9, Av11, Av12, Av14 isolated from alkaloid layer inhibit more apparentely above cells compared to 5-FU (P < 0.05). IC50 values to AGS,A549 cells of above six alkaloids respectively are 0.464μg/ml and 0.447μg/ml.The thesis also provides an overview of the research findings for the Chinese traditional medicine Aconitum plants home and abroad from 1980. It lists the studies recently on their chemical constituents and bioactivities.
