Objective: MIF (macrophage migration inhibitory factor) is a multifunctional cytokine, which has an important role in inflammation and tumor and is the focus of recent researchs.Previous study has confirmed that MIF was expressed in the bladder epithelium and glandular tissue. Through investigating the expression of MIF ,COX-2 (cyclooxygenase-2) and NGF (nerve growth factor) in prostate, bladder and urethra of chemical prostatitis rat models and these organs'inflammatory situation, we explore whether inflammation was induced in the other pelvic organs like baldder,urethra,etc. by chemical prostatitis,and the mechanism of some clinical symptoms of prostatitis.Material and methods:1.20 Rats were randomized to receive injection formalin or saline into the ventral lobe.2.The inflammation condition of the bladder and prostate was verified histologically by HE staining.3.Protein levels expression of MIF,COX-2 and NGF were examined by immunohistochemical and Western blot.Results: Rats injected with formalin mixed with dye showed restriction of the dye to the prostate .Intraprostatic formalin produced inflammation in prostatate;Bladder and urethra inflammation was detected using HE. MIF protein amounts in bladder and urethra decreased, while COX-2 and NGF increased after prostatic injection using immunohistochemistry and western blot.Conclusion: A viscerovisceral reflex in the rat, probably mediated by the lumbosacral spinal cord, produced bladder or urethra hyperreflexia and bladder,urethra edema, and evoked MIF release from the bladder ,urethra and the induction of other inflammatory mediators. Our research try to further confirms the hypothes that MIF is involved in neurogenic inflammation in the pelvic viscera and it may represent an interesting therapeutic target. The inflammation of bladder,urethra might count for some symptoms of prostatitis.
