ObjectiveIn order to solve the burden of aging in the 21st century.This experiment adopted a variety of modern research techniques,observed with sub-acute aged model rats,which treated with D-galactose,in order to explore the regulating function of how important tonifying points work on the central nervous system.Moreover,we compared the difference between conventional electro-acupuncture and the strong electro-acupuncture.As to find a better method in clinical research and supply with further study in basic experiment.MethodsForty three-month-old SD rats,ten of them were randomly selected into a controlled group of normal(group A),and were injected saline in the back skin with a calculate of 125mg/kg·d for forty days,thirty other rats were injected with the same way but the substance is instead of D-galactose.After that,we tested in rats by the filter using water maze.If the time the rat gets to a platform model was significantly increased,or the number of the rat access to blind-side was significant increased,it can account for a successful model. Thirty successful model rats were randomly divided into three groups:model group(group B,ten),conventional electro-acupuncture group(group C,ten), strong electro-acupuncture stimulation.group(group D,ten).Group A and group B was performed with no treatment.In group C,the rats would wear clothing and be acupunctured on Guanyuan and Zusanli in which refer to the "map of the experimental animal acupuncture points",formulated by Acupuncture Research Institute of Experimental of Chinese Acupuncture in 1992.The needle was 15 mm in the length and 0.32mm in the Diameter. Huatuo license,and G6805-Пelectro acupuncture therapy apparatus.We got Guanyuan on negative,while Zusanli on positive,and bilateral Zusanli is in alternate use of the next day,.We selected a continuous-wave,frequency of 2Hz,Current intensity of 1mA,and treated for 30 minutes,once every day,6 times a week,lasting four weeks.At the end of the last treatment,we drawn materials after the overnight.The treating method and drawn materials were the same in group C and D,The difference was that treating time is five minutes and current intensity is 4.5 mA in group D.After ending treatment, we respectively got blood from the eyeball,use radioimmunoassay to detect the quality of serum IL-6.Rats were killed after taking brain tissue,then the tissue is in situ hybridization detection of the expression level of NPY mRNA, with SABC immunohistochemical assay we select the rat IL-6 receptor.At last,we analysis the NPY,IL-6R serum immunoreactive through an Image Analysis System.Results1.Compared with the normal controlled group,the activity of NPY mRNA was lower and the activity of IL-6 and IL-6R was higher in the sub-acute aged model group(P<0.01);2.Compared with the sub-acute aged model group,the activity of NPY mRNA was higher and the activity of IL-6 and IL-6R was lower in the treated group(group C and D)(P<0.05);3.The difference between the conventional electro-acupuncture stimulation treatment group(1 mA) and the strong electro-acupuncture stimulation treatment group(4.5mA) was also significant(P<0.05).What's more,treatment on group D was outstanding. Conclusion1.Acupuncture on important tonifying points can significantly improve aging rats' lower NPY and reduce the high expression levels of IL-6,this may be the impact of brain and central nervous system that enhance the role of immune regulation.2.The effect between conventional electric acupuncture and strong electric acupuncture is significantly different.Though strong stimulation is tolerant,but the efficacy was superior to conventional electricity,and also it lasts shorter,this could provide new ideas in clinical acupuncture for the future.3.This task may explain to provide a kind of scientific basis on neuron immune modulation mechanism at the interpretation of how to improve aging condition when acupunctured on strong point.It also embodied the advantage of the thought that we should protect righteousness as the roof of treatment.
