Currently, the tendency of aging is getting more and more prominent in all of the world. The brain is the most complex and precise argon in human body, and it is also the sensitive one to the aging acting. People pay extensive attention to the issue of brain-aging .It is great practical significance to find an effective medicine to delay the aging of brain, as well as discussing the mechanism of prevention and treatment.Angelica is first recorded in< Shennong ben cao jing >and is regarded as one of the best herb. It is the main medicine with the effect of reliving wind -dampness syndrome. It can regulate and replenish the five viscera—organs especially the kidney, remove blood stasis and resolve phlegm; make the cerebral meridian smooth, therefore, "people can act more quickly and get more healthy after a longer administration". The past research showed that Angelica and its extract with ethanol can improve the flow of cerebral cellular membrane and aging membrane index in the model mice of D-galactose Cerebral aging, intensify the immunity, improve the study and memory ability of the mice .Objective: To observe the effect of Angelica and its extract with ethanol on the Cellular morphology, apopxosis (PCD), genetic control, oxidation injury of cerebral tissue and study and memory function of the natural aging mice , and to discuss the aging function mechanism of Angelica and its extract with ethanol.Methods: take 18-month-old mice as the aging model, test the study and memory with abilities of the mice labyrinth, observe the morphological changes of cerebral tissue under electronic microscope and optical microscope. Test the rate of apoptosis (PCD) and the gene expression of bcl-2, bax through the flow cytometryand immunohistochemical method, test the value of malondialdehyde (MDA) in the brain and the activities of serum superoxide dismutase (SOD) with the method of Thibabituric Acid(TBA)and xanmine oxidase.Results: the study and memory abilities of the natural aging mice descends, the number of neurocyte of hippocampal gyrus decreases significantly, the cells arrange irregularly. The neurocyte of cerebral cortex shows vacuolation degeneration. The early apoptosis/programmed cell death (PCD) and aging changes of cells of neuroglia and neurone can be observed under electronic microscope, including the reduction of nucleus in size, the increasing of Nuclei heterochromatins the deformation of the nucleus, irregular nucleus morphology, the distance of the cellular membrane getting wide: part of mitochondrion crista getting broken. In addition, the rough endoplasmic reticulum (RER) expanding and degranulation, the cytoplasm partly dissolving; lots of lipofuscin depositing. The myelinated nerve fibres of neurocytes. The apoptosis (PCD) of cerebral was high, the gene expression of bcl-2 reduced while that of bax genes increased. The values of cerebral MDA increased, and the activities of plasmatic SOD decreased. Angelica and its extract with ethanol can improve changes that was mentioned above.Moreover, the effect of extract with ethanol shows a better action than that of decoction.Conclusion: Angelica and its extract with ethanol have the function of delaying the aging course of the brain. Angelica and its extract with ethanol may Inhibit apoptosis (PCD) of cerebral cellular cells, probably by controlling oxidization and genes of apoptosis.what's more, make the structure of cerebral histomorphology better, to improve the study and memory ability of mice with aged cerebral cells and delay the aging of cerebrum.
