ObjectiveThe purpose of this study was to investigate the expression of 5-lipoxygenase (5-LOX),vascular endothelial growth factor(VEGF) and microvessel density(MVD) in mucoepidermoid carcinoma(MEC),to explore their relationship and significance in mucoepidermoid carcinoma,to try to provide referential indexes to the treatment and the diagnosis of Mucoepidermoid Carcinoma.MethodsWe have reviewed 40 cases of MEC origining in salivary glands that they were treated at the First Affiliated hospital of Stomatology and oncosurgery of Bengbu medical college during 2004-2008.The expressions of 5-LOX,VEGF and MVD (labeled by anti-human CD34 monoclonal antibody) in 20 cases of normal tissue of salivary glands,20 cases of pleomorphic adenomas and 40 cases of MECs were detected by EliVision TM plus immuniohistochemical method.The correlation of expression of 5-LOX,VEGF and MVD in MECs of salivary glands was analyzed.All data were analyzed by SPSS 13.0 for Windows including Cosstabs x~2 test,Independent samples LSD-T test,Spearman's method,One-Way ANOVA et al.Results1.The positive staining rate of 5-LOX in Mucoepidermoid Carcinoma was 67.5% (27/40),which was significantly higher than in pleomorphic adenoma(0%,0/20) and normal tissue of salivary glands(5.0%,1/20).The expression of 5-LOX differed among MEC and pleomorphic adenomas(P=0.000<0.01),MEC and normal tissue of salivary glands nearby MEC(P=0.000<0.01),pleomorphic adenomas and normal tissue of salivary glands nearby MEC(P=0.311>0.05). 2.The positive staining rate of VEGF in Mucoepidermoid Carcinoma was 65.0% (26/40),which was significantly higher than in pleomorphic adenoma 5.0%(1/20) and normal tissue of salivary glands 10.0%(2/20).The expression of VEGF differed among MEC and pleomorphic adenomas(P=0.000<0.01),MEC and normal tissue of salivary glands nearby MEC(P=0.000<0.01),pleomorphic adenomas and normal tissue of salivary glands nearby MEC(P=0.548>0.05).3.MVD was significantly higher in MEC than in pleomorphic adenoma and normal tissue of salivary glands(24.825±9.685,8.20±1.735,9.9±1.373,P=0.000<0.01). The datas were analyzed by ANOVA(F=51.383,P=0.000<0.001),theywere significantly difference by LSD-t test(P=0.000<0.001).4.The result Of bivariate correlations analysis of Pearson's method showed the expression of 5-LOX and VEGF was positive correlation for MEC(r=0.386,P =0.031<0.05).ConclusionThere wre significant differences in the expression of 5-LOX,VEGF and MVD in MEC,PA and normal salivary glands nearby MEC(P=0.000<0.001).5-LOX,VEGF,CD34 may play an important role in the development of MEC,and may be a useful indexes for clinical treatment and diagnosis.
