The Effect Of Fangxiaoling Capsule On The Apoptosis Of Eosinophile Granulocyte And Tracheal Reconstruction In Lung Tissue Of Bronchial Asthmatic Rats Model

Objective: To investigate the effects of Fangxiaoling capsule on the eosinophile granulocyte(EOS)and tracheal reconstruction in lung tissue of Bronchial Asthmatic Rats Model and explore the mechanism of treating asthma with Fangxiaoling capsule. Methods: Eighty-four male Wistar rats were divided randomly into seven groups: Normal control group, Model control group, Dexamethasone group, Fangxiaoling capsule low dose group, Fangxiaoling capsule middle dose group-1, Fangxiaoling capsule middle dose group-2, Fangxiaoling capsule high dose group, each group had twelve rats. By the way of being sensitized and inspired with OVA , we copied Bronchial Asthmatic Rats Model. At the same time, each group was given relevant medicines. After eight weeks, we killed all the rats. And then we observed the changes of lung histomorphology of rats of each groups, surveyed the rate of apoptosis of EOS and content of gene Fas, gene Bcl-2, TGF-β1,collagen protein of typeⅠ,Ⅲin rats'lung tissue. Results: Changes of bronchial and pulmonary histopathology in each group were typical, it meant that Bronchial Asthmatic Rats Model were successful. Compared to Normal control group, Model control group had more infiltration of EOS, less apoptosis, lower expression of gene Fas, higher expression of gene Bcl-2 and TGF-β1 and more deposit of collagen protein of typeⅢin rats'lung tissue. For the improvement of Asthmatic Rats'general conditions, infiltration of EOS and tracheal reconstruction, Compared comprehensively, Dexamethasone group was better than other groups, Fangxiaoling capsule middle dose group-1 on the verge of it, Fangxiaoling capsule middle dose group-2 and Fangxiaoling capsule high dose group secondly, Fangxiaoling capsule low dose group worstly. Conclusion: Fangxiaoling capsule could alleviate the airway inflammation in bronchial asthmatic rats model. The mechanism of action is that promoting apoptosis of EOS by making gene Fas higher express and gene Bcl-2 lower express. At the same time, alleviating tracheal reconstruction by making TGF-β1 lower express and restaining deposit of collagen protein of typeⅢ. Above-mentioned action can be reinforced by increasing dose or being intervened earlier.
