Research Of Legal Medical DNA Detection By The Domestic Magnetism Beads

Magnetism bead has been used widly, we can use it to extract DNA from many biological samples such as blood,saliva,spermatozoon,perspiration and so on. It operates easily and doesn't need special separate equipment and can avoid losing DNA. In this study we use a domestic Magnetism bead to explore the callback and purification of DNA. The results indicate that the domestic Magnetism bead have high reclaim sensitivity and well purification.,is suitable to apply in Legal medical detection.At first, we use domestic Magnetism bead,silicon beads and IQ kit to extract different quantity standard DNA(9947A), and amplify them by Power Plex16 kit(ABI), 9700(ABI), detect the amplify production by 3100 analyzing instrument(ABI).The results show that Magnetism bead can extract DNA more effectively than silicon beads and IQ kit at PCR of fluorescence ration. Then, we use domestic Magnetism bead,silicon beads and IQ kit to extract the blood which has been diluted 30, 60 times, and also amplify them by Power Plex16 kit(ABI) and 9700(ABI), detect the production by 3100 analyzing instrument(ABI).The results show that Magnetism bead can extract DNA more effectively from samles than silicon beads and IQ kit at trace amount of DNA. Eventually, we use Magnetism bead to extract DNA from three kinds of samples: glass with bloodstain,cells from collar,toothbrush, and amplify them by Power Plex16 kit(ABI), 9700(ABI), detect the production by 3100 analyzing instrument(ABI), all the 16STR loca get the expectable results. Collectively, DNA from polluted samples can be back and purification by domestic Magnetism bead, which recycle DNA from dyestuff, haemachrome, metal ions and the DNA samples can be used to amplify.
