Effects And Mechanisms Of Yifu Preparations On Murine Allergic Contact Dermatitis

Allergic contact dermatitis(ACD) is one of inflammatory dermatitis which is caused by repeated skin and mucous exposure to contact allergens.ACD belongs to delayed type hypersensitivity reaction mediated by T cells. A lot of cytokines, chemotactic factors and costimulatory molecules derived from T cells involved in the whole process of the inflammation. At present, the most of view is that ACD is mediated by Th1cells which secrete IL-2and IFN-γ Imbalance of differentiation between the Thland Th2cells take the responsibility of immune disorders in ACD. The treatment of ACD mainly includes removing allergens,topical corticosteroids,moisturizing and so on Currently,Corticosteroids is the major method to control the acute stage of hypersensitivity reaction and inflammation,But long-term application of corticosteroids will cause systemic side effects,such as adrenocortical insufficiency and secondary infection,and also can cause skin atrophy,angiotelectasis, glucocorticoid-induced cataract, glaucoma, facial erythema, etc,and finally affect children’s growth and adult endocrine system.It is also very common to recur after withdrawal and resist to the treatment. Therefore, combined to the other treatments, looking for effective preparations with less side effects to treat allergic contact dermatitis and reduce cortical steroid dosage are the main task in the clinical work.In this study, Yifu preparations homemade by modern preparation methods were mianly made of Kuh-seng and aloe vera gel, among them the matrine in Kuh-seng has effect of controlling the acute inflammation and has properties of non-steroidal anti-inflammation and immunosuppression,Molecular mechanism research shows that matrine can intervene the expression of chemotactic protein-1(MCP-1) induced by substance P.Aloe vera contains rich mucointin with high polysaccharides, it not only functions as immune regulator, but also helps cell division, and plays an important role in skin care. The preparations have properties of heat-clearing and detoxifying,moisturizing, convergence and anti-itching, and also the preparations are characterized by good curative effect on eczema and dermatitis,convenience for use and no obvious side effects.This study was designed to explore the effects and mechanisms of Yifu preparations on murine allergic contact dermatitis through the establishment of the marine allergic contact dermatitis model. ObjectiveTo observe the Yifu preparation effects and the mechanism of Yifu preparations on murine allergic contact dermatits by establishing marine allergic contact dermatitis model. MethodsThe murine model of ACD induced by2,4-dinitrofluorobenzene(DNFB) was established.ICR mice were randomly divided into6groups-blank control group, DNFB model group,Yifu cream group, Yifu lotion group, Baby eczema cream group and Hydro cortisone Butyrate group.Murine ear swelling and histopathologies were observed. the serum level of IL-2、IFN-γ and the expression of IL-2and IFN-γ in keratinocytes were detected by the method of ELISA and immunohistochemistry respectively. ResultThe results showed murine ear swelling and the expressions of IL-2and IFN-γ in serum or skin lesions were markedly suppressed or down-regulated by either Yifu cream or Yifu lotion or baby eczema cream and Hydrocortisone Butyrate.There was no difference of the effects between Yifu cream and Hydrocortisone Butyrate(P>0.05).But compared with the Yifu lotion and baby eczema cream group, Yifu cream could significantly reducing the ear swelling and down-regulating the expression of IL-2and IFN-γ(P<0.05). Conclusion:Yifu preparations could relieve the inflammation of ACD, the mechanism of its action may be related with inhibiting the release of interleukin-2and interferon-γ. Significance:This study provided the experimental evidence for the development and clinical application of this medicine and verified feasibility of the medicine for treatment of ACD and eczema.The study will play very important roles on removing the patients’sufferings, improveing their quality of life and augmenting the dominant role of traditional Chinese medicine...
