ObjectiveTo investigate the therapeutic effect of SST on the damaged bowel barrier in 7-day-old-SD rats with HIBD,and to explore a new method for the treatment of the barrier damage associated with HIE.Methods1.Establishment of HIBD model:Neonatal 7-day-old SD rats were subjuected to middle anterior neck incision.Left common carotid artery was isolated and ligated, then the rats were exposed to a hypoxic environment(8%02,92%N2)for 2 hours, and the behavior of the rats was observed during hypoxic period.2.Treatment of HIBD model:After 10 minutes,lug/ml of SST diluted with normal saline solution was injected to treatment group intraperitoneally.Seize the rats with left hand,and pierce skin in left or right lower abdomen with right hand.Needle went forward 0.5 to 1.0cm,then formed a 45°angle through abdominal muscles.After fixed,taking the slow injection,the dosage was 20ug/kg.3.Observing behavior of rats:The turnover ability and levorotary ability when their tails were gripped were observed before and after HI.4.General examination of the brain and the ileum:Observed the general appearance of brain and ileum.5.Blood concentration of D-lactic acid,the change in intestinal villus height and width,determination of SP in ileal tissue:The rats were randomly divided into blank group(n=6),sham group(n=6),HIBD model group(n=8),SST-treated group(n =8).After 6 hours,blood was collected from abdominal aorta to measure D-lactic acid,ileum was taken for pathological analysis and determination of SPResults1.Changes of behavior of the rats:Before the experiment all rats were healthy. After HI,the rats in sham group were still healthy,while 9 rats couldn't turn themselves over and 2 rats became levorotary when their tails were gripped in the 16 rats which bared HI injury,There were 3 rats exhibited both behavior problems.2.General examination of ileum:Compared with blank group,model group showed obvious pallor and edema after 6 hours.Treated group showed no significant change or little edema.Sham group showed no change.3.General examination of brain:Ligated brain hemisphere of model group showed obvious pallor and edema,and showed liquefactive necrosis in one of the rats.Treated group showed no significant change or little edema.The two brain hemispheres of sham group showed no pathological change.4.D-lactic acid blood concentration,intestinal villus height and width changes determination of ileal tissue SP:In model group the level of D-lactic acid obviously increased.Cillia of ileum became widen and shorten,the number decreased.And in stratum mucosa and neurofibrillary tangle,the expression of SP increased.Compared the level of D-lactic acid decreased in treated group,intestinal villi became thinner and higher,even the number increased.Besides that,the expression of SP decreasedConclusions1.The HIBD model was successfully established.2.:In model group the level of D-lactic acid obviously increased.Cillia became widen and shorten,and the number decreased.At the same time in stratum mucosa and neurofibrillary tangle,the expression of SP increased,which suggested that the bowel barrier damage of HIBD maybe related to inflammation.3.The level of D-lactic acid decreased in treated group,intestinal villi became thinner and higher,the number increased,and the expression of SP decreased.SST showed therapeutic effect on bowel barrier damage of HIBD are sure.Our result opens a new window for the treatment of bowel barrier damage associted with HIE.
