Objective:To detect the expression of E-cadherin,β-catenin and CD44v6 in gastric carcinomas and lymph-node metastatic tumors,and to probe the relationship between their expression and the differentiation,depth of invasion and lymph node metastasis of the gastric carcinomas.To investigate the effect of E-cadherin,β-catenin and CD44v6 on carcinogenesis,progression,invasion and metastasis in gastric carcinoma, which provided theoretical evidence for early diagnosis,rational therapy and prognosis judgment.Methods:70 cases of gastric carcinoma specimen,with lymph node metastasis 48 cases and normal gastric tissue 28 cases were obtained from patients who had undergone surgical operations of gastric cancer in Affiliated hospital of Hainan Medical college and the people's hospital of Haikou between October 2005 and December 2007.The lymph nodes were detected by clearing fat method.The expression of E-cadherin,β-catenin and CD44v6 in all cases were detected by immunohistochemistry staining.Results:1.Immunohistochemical study showed that E-cadherin was expressed in 25 cases of normal gastric tissue,46 cases of gastric carcinoma tissue and 22 cases of lymph-node metastatic tumors,with positive rate of 89.29%,65.71%,and 45.83%, respectively.The expression of E-cadherin in gastric carcinoma was associated with the depth of invasion and lymph node metastasis.It was unrelated with the carcinoma differentiation.2.Immunohistochemical study showed thatβ-catenin was expressed in 26 cases of normal gastric tissue,48 cases of gastric carcinoma tissue and 27 cases of lymph-node metastatic tumors,with positive rate of 92.86%,68.57%,and 56.25%, respectively.The expression ofβ-catenin in gastric carcinoma was associated with differentiation,the depth of invasion and lymph node metastasis.3.Immunohistochemical study showed that CD44v6 was expressed in 8 cases of normal gastric tissue,50 cases of gastric carcinoma tissue and 40 cases of lymph-node metastatic tumors,with positive rate of 28.57%,71.43%,and 83.33%, respectively.The expression of CD44v6 in gastric carcinoma was associated with the depth of invasion and lymph node metastasis.It was unrelated with the carcinoma differentiation.4.The expression E-cadherin and CD44v6 was negatively associated in gastric carcinoma tissues(χ~2=14.609,P=0.000,r=-0.457).There was non-statistical correlation between E-cadherin andβ-catenin orβ-catenin and CD44v6 expression in gastric carcinoma.5.In lymph node metastatic tumor tissues,the expression of E-cadherin and CD44v6 was negatively associated(χ~2=8.878,P=0.003,r=-0.486),whereas,the expression of E-cadherin andβ-catenin was positively associated(χ~2=7.294,P=0.007,r =0.390), and there was non-statistical correlation on expression ofβ-catenin and CD44v6 (χ~2=0.610,P=0.435,r=-0.169).Conclusions:1.E-cadherin was over expressed in normal gastric tissue and obviously low expression in gastric carcinoma and lymph node metastatic tumor tissue,which showed that the expression of E-cadherin in gastric carcinoma was associated with the depth of invasion and lymph node metastasis,unrelated with differentiation.It indicated that E-cadherin showing the inhibition effect on progression,invasion and metastasis in gastric carcinoma.2.β-catenin was over expressed in normal gastric tissue and obviously low expression in gastric carcinoma and lymph node metastatic tumor tissues,which showed that the expression ofβ-catenin in gastric carcinoma was associated with differentiation,the depth of invasion and lymph node metastasis.It indicated thatβ-catenin involved in the carcinogenesis of gastric cancer,progression,invasion and metastasis.3.CD44v6 was lower expressed in normal gastric tissue and obviously high expression in gastric carcinoma and lymph node metastatic tumor tissues,which showed that the expression of CD44v6 in gastric carcinoma was associated with the depth of invasion and lymph node metastasis.It was unrelated with the carcinoma differentiation.It indicated that CD44v6 may have promotive effect on the progression,invasion and metastasis of gastric carcinoma.CD44v6 could be used as an indicator of invasion and metastasis in gastric carcinoma.4.The expression E-cadherin and CD44v6 was negatively associated in gastric carcinoma tissues.There was non-statistical correlation between E-cadherin andβ-catenin orβ-catenin and CD44v6 expression in gastric carcinoma,which indicated that E-cadherin and CD44v6 has the relation of mutual constraints during the carcinogenesis of gastric carcinoma.5.In lymph node metastatic tumor tissues,the expression of E-cadherin and CD44v6 was negatively associated,whereas,the expression of E-cadherin andβ-catenin was positively associated,and there was non-statistical correlation on expression ofβ-catenin and CD44v6,which indicated that E-cadherin and CD44v6 has the relation of mutual constraints and promote each other between E-cadherin andβ-catenin during the invasion,metastasis of gastric carcinoma.
