Preliminary Study On Flavonoids,Volatile Oil,Microelements Of Diaphragma Of Juglans Regia L.

The major purpose of this paper is that do some research on flavonoid,volatile oil and microelement of the diaphragma of Juglans regial L., and that can provide theory evidence and reference for the development of food and the pharmaceutical industry .The major contents and results are as follows:It was proved that flavonoid was exist in the diaphragma of Juglans regial L.with experiments .To extracted the total flavonoids of the diaphragma of Juglans regial L. with conventional heating reflux extraction and ultrasound-assisted extraction method.To analyze the total content of flavonoids in diaphragma of Juglans regial L.by spectrophotometry.Rutin was used as reference and the detecting wave length was 510 nm.It was provided that ultrasound-assisted extraction is better than conventional heating reflux extraction because the yield of total flavonoids was higher with ultrasound-assisted extraction and it can save time and solvent.Response Surface Analysis(RSA) was applied to optimize the extraction of the total flavonoids of diaphragma of Juglans regial L. Ethanol concentration, time and expects the fluid to compare were chosen as the factors in RSA . The optimum condition obtained from RSA was as follow: the ethanol concentration 57%, time 30min, expects the fluid to compare 1:25.Flavonoid in the diaphragma of Juglans regial L. was purified with the AB-8 pocket adsorption resin, the adsorption rate was 89.51%, the desorption rate was 73.47%, purity was 23.4%.The method of measuring antibacterial activity is proceeded for flavonoid in different solvents, such as 60% ethanol and ethyl acetate.The results showed that ethyl acetate extract from diaphragma of Juglans regial L. had no antibacterial activity on this four kinds common bacteria and the bacteriostasis of 60% ethanol extract was not obvious. The volatile oil extracted from diaphragma of Juglans regial L. was analysed by gas chromatography-mass spectrometry. The result showed that 11 compounds was exist.The microelements of diaphragma of Juglans regial L.was determined by flames atom absorb.Ten kinds of elements was detected as follows: Na,K,Ca,Mg,Zn,Fe,Cu,Ni,Co,Mn.
