Bidens bipinnata L. is a species of the genus Bidens (Compositae). It is widely distributed in majority province in China. Bidens bipinnata L. had been traditionally used for many years. It is bitter in taste, neutral in nature and innocuity, and has the effect of heat-clearing, detoxicating and eliminating stasis to subdue swelling. It contains many chemical compositions, such as flavonoid glycoside, saponin, alkaloid, aetherolea. It was largely used to treat dysentery, diarrhea, hepatitis, stomachache. The quality standard of Bidens bipinnata L. has not been reported. In order to develop a new use of Bidens bipinnata L., the quality standard and fingerprint of Bidens bipinnata L. has been studied in this paper. The main contents were divided into two sections as follows:1. The quality standard of Bidens Bipinnata L.This experiment research into the quality standard control of Bidens bipinnata L. and establish the methods of quality estimate of Bidens bipinnata L .The routine methods was used of pharmacognosy such as freehand section and surface slice to observe the microscopic characters of the leaves and stems of Bidens bipinnata L .and thin layer chromatography (TLC) to identify it. The results showed the plant is Bidens biternata (Lour.)Merr.et Scherff. According to technical requirements in the appendix in China Pharmacopoeia (Ⅰ, 2005 ED).The water content of 8.15~9.16%, total ash content of 6.72~8.49% and acid endurable ashes content of 1.06~1.27% was obtained. Organochlorine pesticide residues in Bidens bipinnata L. has been determination by using gas chromatography. BHC in crude drug is 3.7×10-8(g/g). Meanwhile, we apply colorimetric method to determine the content of TFB. The average contents of TFB was 7.41%. We also use HPLC to detect the content of hyperoside. The average content of hyperoside was 0.049%.2. The fingerprint of Bidens Bipinnata L.The fingerprint method based on high-performance liquid chromatography for the quality control of Bidens bipinnata L were firstly described and optimized. HPLC system was used to obtain the fingerprint spectrum of Bidens bipinnata L. The chromatography condition was ODS C18 column(4.6 mm×255 mm,25μm),mobile phase Methanol-0.2%HAc,and flow speed 1.0 mL·min-1, detection wavelength 363 nm.17 peaks ascertained as the common peaks of the fingerprint form its unique fingerprint. Comparing with the reference of hyperoside, the relative preserving time, the relative areas of other peaks and the similitude degree of each fingerprint are calculated as the technique parameters to estimate the fingerprint. In this experiment, the standard fingerprint has been established and the similitude degree of 10 batches Bidens bipinnata L. has also been obtained (0.941~0.999).The facts mentioned aboved illuminate that these experiments successfully build the quality standard of Bidens bipinnata L and the FT of Bidens bipinnata L by HPLC. The method is accurate, reliable, good reproducibility, strong characteristic and can be used for the standard of genera judgement and the quality estimation about the Bidens bipinnata L. .
