Object: The purpose of this study was to ivestigate the expression ofα-smooth muscle action(α-SMA),peroxisome proliferators-activated receptorγ(PPAR-γ), detect the proliferation , apoptosis and the amount of triglyceride on rat HSC-T6 under the control of Quercetin, to find out the effects of Quercetin on hepatic fibrosis.Methods: Used passage HSC-T6 as study objects. The experiment was divided into 5 groups: The first group was blank; the second group from group 2 to group 5, different amounts of Quercetin was added to the activated HSC-T6, to made the concentration of Quercetin to be 10,20,40,80μmol/L respectively. After each group was incubated for 24 hours, observed the apoptosis under AOEB dye. MTT—assay was used to observe the inhibitory effect of quercetin on HSC-T6;Detected the intracellular amount of total trigelyceride through high performance liquid chromatography (HPLC) .Theα-SMA, PPAR-γexpression of the HSC-T6 of each group was detected by RT- PCR respectively ,thereby make out the amount– effect relation curve. And exposed the cell to Quercetin at the best concentration for 12 h,24 h,48 h respectively. Then detected theα-SMA, PPAR-γmRNA's expression of the HSC-T6, and draw out the duration-effect relation curve. Using±s to represent experimental data, using SPSS13.0 statistical software to analyze the data, a P <0.05 was considered significantly important.Results: The result displays: Quercetin increased the intracellular trigelyceride of the HSC-T6. What's more,it decreased theα-SMA mRNA expression and increased the PPAR-γmRNA expression concentrationally dependently at the same time; Then After mixing the Quercetin of 40μmol/L with the HSC-T6 for 12,24,48 hours, the result says: the increase of trigelyceride in the cells by Quercetin was dependent on the duration , at the same time the decrease ofα-SMA and increase of PPAR-γmRNA's expression was dependent on the duration dependently.Conclusions:1. Quercetin could increase apoptosis and decrease activation, proliferation of HSC-T6.2. Quercetin can increase the quantity of trigelyceride in the activated HSC-T6; Quercetin can reverse the activated HSC-T6; Quercetin could upregulation the PPAR-γmRNA in HSC-T6...
