Objects: Our study was designed to construct the rat model with chronic atrophic gastritis (CAG), and observed the preventive effect of lipoxygenase inhibitor nordihydroguaiaretic acid (NDGA) on CAG rats, then try to investigate the molecular mechanism and to provide theoretical evidence for the chemical prophylaxis of chronic atrophic gastritis .Methods: 72 male rats (6-week) were randomly divided into 6 groups: normal group; CAG model group; NDGA group with different dose (10mg/Kg, 20mg/Kg, 30mg/Kg which through hypo in abdominal wall Qod after the 18th week of experiment); positive group (folacin). All groups were freely drink 100μg/ml MNNG expect normal group. Rats' consciousness, appetite, glossiness of clothing hair, sedes and body weight were observed during experimental period. 24 weeks later, all rats were executed, their stomach mucous membrane were viewed by HE staining; The expression of 5-LOX and P16 protein in mucous membrane epithelioid cells were detected by immunohistochemistry. Data from each experiments were expressed as mean±SD. Measurement data were analyzed by F test and enumeration data were byχ2 test by using SPSS 13.0 software, P<0.05was considered significant.Results: (1)There were 66 rats remaining after 24 weeks' feeding. The CAG incidence rate in the model group was significant higher than normal group (P<0.05), indicated that the model was established successfully. (2)The body weight of model rat was lower than normal group,NDGA and folacin group (P<0.05), there is no significant difference between other groups (P>0.05). (3)In the aspect of prevention, NDGA groups and positive group could prevent CAG (P<0.05), but there was no significant difference between various NDGA groups and positive group (p>0.05). (4)The expression level of 5-LOX and p16 protein were different between model and NDGA groups (P<0.05), but there is no statistical significance between NDGA and folacin group.Conclusions:1. NDGA could prevente the occurrence of N-ethyl-N-nitro-N-nitrosoguanidine-induced chronic atrophic gastritis in rat;2. NDGA could down-regulate the expression of 5-LOX and up-regulate the expression of P16 in stomach mucous membrane of N-ethyl-N-nitro -N-nitrosoguanidine-induced chronic atrophic gastritis in rat.
