Objective:To investigate and compare the prophylaxis effect of Radix Salviae Miltiorrhizae and pravastatin on steroid induced Avascular Necrosis of Femoral Head(SANFH).Methods:46 Japanese white rabbits were randomly divided into five groups:Normal group (GroupA),Model group(GroupB),Radix Salviae Miltiorrhizae group(GroupC),Pravastatin group(GroupD),Combined use of Radix Salviae Miltiorrhizae and prophylaxis group(GroupE).The animal model was made by the method of Matsui.B,C,D,E group were injected 10ml/kg of sterile horse serum intravenously two times at three-week intervals,When the last time injection serum equinum later,interval 24 hour,40mg/kg/day of methylprednisolone acetate were injected intraperitoneally for three consecutive days.After received the methylprednisolone acetate,they were taken the medicine.B group have the 2ml saline every day.C group were received 0.5ml/kg/day Radix Salviae Miltiorrhizae.D group were received 1.2ml/kg/day Pravastatin.A group were given normal animal feeds.E group was received Radix Salviae Miltiorrhiza and Pravastatin.Indexes of hemorrheology,fatty metabolism,X-rays and the Histological observation of femoral head were Carried out five week later.To acquire the relevant information and data.Results:①Detection of the blood fat:serum levels of TC,TG and LDL in B group were significantly higher than those in other groups,and such markers decreased in E,D and C group(P<0.01-P<0.05)。②Measurement hemorrheology:whole blood viscosity,plasma viscosity,the exponent of Erythrocyte aggregation in group B increased significantly compared with those in other goups,and E,D and C groups showed smaller values of the above indexes(P<0.01-P<0.05 )。③X-rays examination:There were no any collapse in the articular surface of femoral head.The group B of circumscription femoral head is no clear.It had some chang.we can find that it was cystis degeneration and sclerosis in the most of femoral head.Chose chang were not exit in other groups.④Histopathology watch:The group A is normal.Bone trabecular of femoral head in E,D and C relatively B thick,athe texture more regulation and abundant red blood corpuscles;The empty bone lacuna of B group increases obviously than E,D and C.It had significant differentces(P<0.05).The histopathology of group E was likely the group A.⑤Ultrastructure watch:In the C,D and Egroup,osteocytes is abundant,nucles is relatively normal,chromatins is homogeneous,organelles is abundant and there are many and clear collagenous fibrils;In the B group,osteocytes is relatively little and it is obvious more cell dwindle by volume,pyknotic nudes,marginal chromatins,the organelles is reduce,swelling mitochondria increases,the collagenous fibrils is sparse.E group approaches Agroup.Conclusions.1,We use the method of Masui and make the animal model of steroid induced avasacular necrosis of femoral head in the early.This animal model is suitable for being used in studying the stetoid induced necrosis of the femoral head.2,The Radix Salviae Miltiorrhizae can improve hemorrheology obviously in the rabbit model of SANFH.It can reduce the empty lacuna in the femoral head.So it can prevent the SANFH.3,Pravastatin was able to improve the hyperlipoidemia in the SANFH,It also can reduce the empty lacuna,it can prevent the SANFH.4,Combined use of Radix Salviae Miltiorrhizae and prophylaxis is more useful than only one for prevent the SANFH.
