1 Objective:To investigate the anti-proliferation of Quercetin or 17-AAG on human Uterine Cervix Cancer HeLa cells and chinese hamster fibroblast cells;to evalute the enhancement of radiosensitivity by Quercetin or 17-AAG on the two kinds of cells.2 Mehtods:HeLa cells and V79 cells were cultured in differect concentrations of Quercetin or 17-AAG in vitro.The inhibitory ratio of the cells were measured by MTT assay.Colon forming assay is carried out to study the enhancement of radiosensitivity by Quercetin or 17-AAG on HeLa cells and V79 cells.3 Results:1) Quercetin could inhibit the growth of the HeLa cells and V79cells in vitro significantly .The suppression was both in the dose- dependent and time-dependent.2) Colon forming assay showed Quercetin definitly enhanced radiosensitivity of the HeLa cells and V79 cells. The average deadly dosage(D0)of the cells has decreased ,the Dq value is less than that in the sole irradiation group and the surviving curve Dq becames smaller.When the Quercetin concentration is 150μmol/L, the sensitization enhancement ratio (SER)is approximation to 2.0 and 1.5.3) MTT result showed the inhibition of 17-AAG on the HeLa cells was in the dose-dependent and time-dependent,and 17-AAG had less effect on the proliferation of V79 cells.4) To observe the radiosensitizing effect of 17-AAG on the HeLa cells and V79 cells in clone formation assay, 17-AAG enhanced the radiosensitivity of the tumor cells. The average deadly dosage(D0)of the cells has decreased ,the Dq value is less than that in the sole irradiation group and the surviving curve Dq becames smaller.When the 17-AAG concentration is 75nmol/L, the sensitization enhancement ratio (SER)is approximation to 2.3.17-AAG had no effect on the radiosensitivity of V79 cells.4 Conclusions:Quercetin could inhibit the growth and enhanceand the radiosensitivity of the HeLa cells and V79 cells,17-AAG could inhibit the growth and enhanceand the radiosensitivity of the HeLa cells , in contrast with, 17-AAG had less effect on the radiosensitivity of V79 cells.
