To investigate the quercetin's effect on proliferation and radiosensitivity of blood lymphocyte and bone marrow nucleated cells in rats.Mehtods:Using CCK-8 method and Annexin V/PI method to study proliferation inhibition and apoptosis of the impact and effects of radiosensitivity and to explore its possible mechanism of quercetin on Raji Burkitt's lymphoma cell. Using CCK-8 method to study the effect of quercetin and 17-AAG on proliferation and radiosensitivity of the rat blood lymphocytes and bone marrow cell . Using CCK-8 method to study the effect of 17-AAG combined quercetin on cell proliferation of the rat blood lymphocytes.Results : 1) Quercetin could inhibit the growth of the Raji cells in vitro significantly .The suppression was both in the dose- dependent and time-dependent. And quercetin definitly enhanced radiosensitivity of the Raji cells.2) Quercetin have a protective effect on blood lymphocyte and bone marrow nucleated cells in rats, and quercetin definitly diminished radiosensitivity of the blood lymphocyte and bone marrow nucleated cells in rats.3) CCK-8 assay showed 17-AAG have a significantly inhibited effect on blood lymphocyte cells in rats. The suppression was both in the dose- dependent and time-dependent. And 17-AAG did not affect radiosensitivity of the blood lymphocyte cells in rats. 17-AAG have a significantly inhibited effect on bone marrow nucleated cells in rats. The suppression was in the dose-dependent. And 17-AAG definitly enhanced radiosensitivity of the bone marrow nucleated cells in rats. 4) CCK-8 assay showed different concentrations of 17-AAG combined quercetin on the rat blood lymphocytes have some synergistic inhibition effect.Conclusions:The results show that quercetin could inhibit the growth of the tumour cell in vitro significantly. And quercetin enhanced radiosensitivity of the tumour cell. Quercetin have a protective effect on Normal cells, and quercetin diminished radiosensitivity of Normal cells. 17-AAG have a inhibited effect on Normal cells. And 17-AAG did not affect radiosensitivity of the Normal cells. Different concentrations of 17-AAG combined quercetin on the rat blood lymphocytes have some synergistic inhibition effect.
