Objective To study the effects of Xuefuzhuyu extractive in acute bacterial peritonitis rats on the concentration of NO and endotoxin in plasma,TNF-αand IL-10 in serum;the bacteria translocation with mesenteric lympha node and liver and the expression of NF-κB in intestinal mucosa.Methods 50 Sprague-Dawley rats were intraperitoneally injected with Escherichia coli.to induce acute bacterial peritonitis;meanwhile,Xuefuzhuyu extractive water-solution of 10,15 and 20 times the adult dosage was intragastricly given to the low(L),medium(M),high(H) therapeutic groups individually.After 66 hours,blood was collected from angular venous plexus and kept for measuring the plasmatic NO with the method of nitrifying ferment and endotoxin with LAL(Limulus Amebocyte Lysate).After 72 hours,all animals were sacrificed,blood and tissues were sterily collected for measuring the serous TNF-αand IL-10 with ELISA(Enzyme Linked Immunosorbent Assay),the bacteria translocation with mesenteric lympha node and liver with the mothod of bacterial culture,and the expression of NF-κB in intestinal mucosa with immunohistochemical staining.Results The concentration of endotoxin and the expression of NF-κB in intestinal mucosa in therapeutic groups was lower than model control(C)group(P<0.05 or P<0.01);the concentration of TNF-αand the bacteria translocation with liver in L group is lower than C group;there was no significant difference between C group and therapeutic groups in the concentration of IL-10 and the bacteria translocation with mesenteric lympha node,but the concentration of NO was significantly higher in M and H groups than in N group(P<0.01).Conclusion The Xuefuzhuyu extractive can play an important role in reducing the concentration of endotoxin in plasma and TNF-αin serum,decreasing the incidence of bacterial translocation,and inhibiting activation of NF-κB in intestinal mucosa.
