Objective: 1)we detected the expression of survivin in cervical precancerous lesion and invasive uterine cervical cancer, to determine whether Survivin expression was related to in the critical steps of cancer onset and progression, and evaluated its diagnostic value in cervical cancer. 2)To investigate the expression of VEGF,EGFR in cervical carcinoma and premalignant lesion, and explore their role in the pathogenesisi of cecical cancinomal. Method : Immunohistochemistry staining and Real- time fluorescence quantitative PCR were used to detect the expression of survivin in 30 invasive cervical cancer,15 cervical precancerous lesion and 10 cervicitis tissue. Rerults: 1)Survivin expression tended to increase along with tumor progression-from cervical precancerous lesion to cervicitis lesion, with a statistical significance ,had most expression in invasive cervical precancerous lesion but only a low expression in cervicitis tissue.(P<0.05å’ŒP<0.01). 2)Contrast to cervicitis tissue, SurvivinmRNA expression was elevated in cervical precancerous lesion and invasive uterine cervical cancer(P<0.05å’ŒP<0.01), add up to 11.0 and 22.6 fold. 3)SurvivinmRNA was positively correlated with survivin protain in cervical cancer,cervical precancerous lesion(p=0.02å’Œp=0.01),but no correlation was fond in cervictis. Conclusion:1)Survivin significantly related to the genesis and progression of cervical cancer, and was an important tumor marker. Real-time fluorescence quantitative PCR used to detect the expression of Survivin could be a reliable way for early diagnosis of cervical cancer.2)The quantitative RT-PCR approach used in our study may be sensitive enough to detect the differences of survivnmRNA , and it provide a rationale approach for the diagnosis of cervical cancer. 3)We hypothesize that VEGF,EGFR will up-regulate survivin protain and survivin mRNA, pay an important role in the same molecular mechanism.
