| Purpose:To study the chang of the expression of IGF-I, IGFBP-1, IGF-IR and the endometrial epithelial cell's growth state in the mid-secretory Endometrial epithelial cell of normal and PCOS combined IR/HI simulated by Ishikawa cell. And searching for the reasons of the low embryo implantation rate and high spontaneous preganancy loss rate of PCOS patients is to improve the clinical pregnancy rate in patients with polycystic ovary syndrome.Method:Addedβ-ESTRADIOL (E2) 200pg / mL and progesterone (P) 20ng / mL to simulate the endometrial mid-secretory. Simultaneously, this group would be respectively treated with 0μU/mL,10μU/mL, 30μU/mL ,100μU/mL insulin and then the group were divided into the negative control group, control group, experimental group and the positive control group. Ishikawa cells were cultured in DMEM with 10% FCS for 24 hours.After 24 h of culturing ,used RT-PCR method to extract the IGF-I, IGFBP-1, IGF-IR mRNA expression and the MTT method to observe the cells'growth.Results:1.The expressions of IGF-I mRNA, IGFBP-1 mRNA, IGF-IR mRNA were positive in the 4 group. The comparison of IGF-I mRNA, IGFBP-1 mRNA expression in the 4 group were more significantly (P <0.001, P <0.0001). The IGF-I mRNA expression increased and IGFBP-1 mRNA expression decreased after treated with insulin ,in a dose-manner,in Ishikawa cells.2.IGF-I mRNA in the experimental group expressed higher then the control group. IGFBP-1 mRNA in the experimental group expressed lower then the control group. (P<0.0001,P<0.0001), There is no significantly change of IGF-IR mRNA in the both two group (P>0.05).3.The change of the growth of the Ishikawa cells were more significantly (P <0.005) The cells'growth increased in a dose-manner.4.The cell growth and the IGF-I mRNA in the experimental group were positive correlated. (r=0.672, P<0.001). The cell growth and the IGFBP-1 mRNA in the experimental group were negative correlated (r=-0.618,P<0.001).Conclusion:1.There is a chang of the expression of IGF-I, IGFBP-1 in the mid-secretory endometrial cell between normal and PCOS combined IR/HI patients.And this may be one of reason for the low embryo implantation rate of PCOS patients.2.There is a chang of the endometrial epithelial cell's growth state in mid-secretory between normal and PCOS combined IR/HI patients.And this may be another one of reason for the low embryo implantation rate of PCOS patients.3.The mutual influencence between the change of the expression of IGF-I, IGFBP-1 in the mid-secretory endometrial cell and the endometrial epithelial cell's growth state in PCOS combined IR/HI patients can produce the mid-secretory endometrial cell gene abnormal expression increased and cell proliferation and apoptosis disorders, further affect embryos implantation in patients with PCOS. |
