Objection:Through the establishment of the experimental rat model of diabetes and giving fluvastatin treatment intervention, observing the expression of Tumor necrosis factor (TNF) in rats blood and urine and the renal disease, investigating the effect of TNF-αon diabetic nephropathy and the mechanism of statins pretect the kidney, providing a theoretical basis for the prevention and treatment of diabetic.Methods:The male SD rats which provided by the experimental animal center of Medicine College of NanChang University, weighing 180-220g, were randomly divided into normal control group(group A), diabetes mellitus model control group (group B), diabetes mellitus model for the intervention group (group C). Inject STZ into abdomen of group A and group B to establish diabetic rats model, and After oral administration of fluvastatin to group C for 8 weeks, the results were analyzed in blood sugar, blood fat, urine protein, renal function and the expression of TNF-αin serum and renal of each group.Results:(1)Blood sugar: group B(26.11±6.53mmol/L)and group C(22.98±4.58 mmol/L)were still significantly higher than group A (9.90±2.09 mmol/L)(P<0.01)after 8 weeks. The experimental rat model of diabetic was successfully established and no spontaneous remission, and there was no significant differences in blood glucose between group B and group C(P> 0.10).(2)Lipid: Triglyceride levels and Total cholesterol in rats among the three groups were no significant differences (p>0.10).(3)Renal function: there was no significant difference among the three groups in Serum Creatinine (p>0.10); but group B (15.46±2.53 mmol / L) and group C (12.58±2.01 mmol / L) were significantly higher than group A (10.08±2.03 mmol / L) (P <0.05) in Blood Urea nitrogen (4)Urinary protein/Creatinine ratio: both group B(0.185±0.048) and group C(0.105±0.021) were significantly higher than group A ( 0.031±0.006 )(P<0.01),while group C was lower than group B(p<0.01).(5)Serum TNF-α: group B (2.23±0.49) and group C (1.64±0.29) were significantly higher than group A(1.04±0.21)in serum TNF-α(P<0.01), while group C was lower than group B(p<0.05).(6)Renal TNF-αexpression by immunohistochemistry: there was no or little TNF-αexpression in the glomerular and tubulointerstitial of group A (3.71±2.430), but had a strong expression in the glomerular mesangial area of group B (32.00±5.182) (P<0.01). The expression of TNF-αalso expressed in mesangial areal of group C (20.62±5.605), but weaker than group B(p<0.01).Conclusion:Fluvastatin can inhibit the expression of TNF-α, so as to reduce the excretion of urine protein, mitigate the pathological changes of renal, retard the development of diabetic nephropathy.
