Object:To observe the influence of different concentration gradient neuromedion B receptor antagonist on gestational age and delivery interval of pregnant mice.Study the mechanism of nuclea factorκB—heat shock protein 70,interlukin-6 passway participating in the labor on set.Methods:1.Establish patterns of pregnant mice the gestational age of which is precise and divide them into five groups randomly which are low-dose I123 group,middle-dose I123 group and high-dose I123 group and positive contrast ritodrine group and negative contrast physiological saline group was respectively injected by i123 50μg/kg,150μg/kg,250μg/kg,2mg/kg and equal amount physiological saline.2.Observe the influence of different grade doses NMBR antagonist on gestational age and delivery interval of Mus musculus.3.Harnessing the analytical method of transcription factor Noshift detects the influence of different grade doses NMBR antagonist on binding activity of NF-κB—P65 DNA in myomitrium of pregnant mice.4.Using semi-quantitative RT-PCR and technology Western blotting measures the influence of different grade doses NMBR antagonist on mRNA and protein expression level of IL-6 and HSP70 in myomitrium of pregnant mice.Results:1.The influence of different grade doses NMBR antagonist on gestational age and delivery interval of pregnant mice.1) The gestational age of negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group was respectively 19.44±0.82,19.38±0.25,19.60±0.61,19.26±0.34 and 19.47±0.68 days.Clearly seen that,middle-dose I123 group give the longest time of gestation.Compared with the five groups,they don't have notable difference(P>0.05).2) The modified gestational age of low-dose I123 group, middle-dose I123 group and high-dose I123 group,positive contrast ritodrine group and negative contrast physiological saline group is respectively 10.50±19.75,9.00±6.02,14.45±14.71,6.33±8.06 and 14.15±21.90 hours.The modified gestational age of middle-dose I123 group give longest time.Compared with the five groups,they don't have notable difference(P>0.05).3) The delivery interval of negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group is respectively 16.60±5.85, 13.00±4.90,26.70±13.99,19.25±10.47 and 22.00±10.20 minutes.Clearly seen that,middle-dose I123 group give the longest time of the delivery interval.Compared with low-dose I123 group and negative contrast physiological saline group,it have statistical significance(P<0.05)Comparing the delivery interval of pregnant mice with high-dose I123 group and positive contrast ritodrine group,we find they don't have notable difference(P>0.05).2.The mechanism about dilevery interval of pregnant mice affected by antagon NMBR with different concentration gradient.1) the expression of binding activity of NF-κB—P65 DNA in myomitrium of pregnant mice.Negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group is respectively 0.112±0.451, 0.088±0.026,0.146±0.061,0.102±0.041 and 0.114±0.036.The expression level of middle-dose I123 group is highest.Compared with negative contrast physiological saline group and positive contrast ritodrine group,we find it doesn't have notable difference(P>0.05).Then Comparing the expression of binding activity of NF-κB—P65 DNA of pregnant mice in three-dose I123 group,we find all of them don't have notable difference(P>0.05).2) the expression of HSP70 mRNA in myomitrium of pregnant mice is respectively 0.308±0.146,0.276±0.284,0.514±0.260,0.658±0.263 and 0.059±0.125 in negative contrast physiological saline group, low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group.The relative expression level of high-dose I123 group is obviously superior to negative contrast physiological saline group,positive contrast ritodrine group and low-dose I123 group(P<0.05);Compared with middle-dose I123 group,we find it doesn't have notable difference(P>0.05).3) the expression of protein HSP70 in myomitrium of pregnant mice is respectively 0.410±0.23,0.195±0.11,0.264±0.15,0.450±0.14 and 0.274±0.060 in negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group.The expression of protein HSPT0 in low-dose I123 group is obviously lower than that of negative contrast physiological saline group and high-dose I123 group,and the differences between them have statistical significance(P<0.05).the expressions of protein HSP70 in myomitrium of pregnant mice among the low-dose I123 group, middle-dose I123 group and positive contrast ritodrine group,we find that it doesn't have notable difference(P>0.05),it also doesn't have notable difference between middle-dose I123 group and high-dose I123 group(P>0.05).4) The expression of IL-6 mRNA in myomitrium of pregnant mice is respectively 0.133±0.184,0.235±0.174,0.279±0.094,0.335±0.194 and 0 in negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group.The relative expression level of high-dose I123 group is obviously superior to negative contrast physiological saline group,positive contrast ritodrine group and low-dose I123 group(P<0.05);Compared with middle-dose I123 group,we find it doesn't have notable difference(P>0.05).5) The expression of protein IL-6 in myomitrium of pregnant mice is respectively 0.052±0.086,0.058±0.096,0.073±0.101,0.024±0.039 and 0.050±0.082 in negative contrast physiological saline group,low-dose I123 group,middle-dose I123 group,high-dose I123 group and positive contrast ritodrine group.The relative expression of protein IL-6 in myomitrium of pregnant mice,the high-dose I123 group is lower than negative contrast physiological saline group and positive contrast ritodrine group,but it doesn't have statistical significance between them(P>0.05).Compared with middle-dose I123 group,it also don't have statistical significance between them(P>0.05).Conclusion:1.NMBR antagonist(I123) doesn't have exetend gestational age.but it can exetend delivery interval.2.The mechanism of NMBR antagonist—I123 exetend delivery interval independent NF-kB—HSP70,IL-6transcription ways.Maybe there are other transcription ways or transcription factor of NF-kB involed in signal transduction pathways.
