| With Sprapue-Dewley rat models with 5/6 nephrectomy , This research observed the effects of YISHEN decoction on the renal function , pathologic changes of the remnant kidney , expression of IV-Col and MMP-2 in the kidney.Results:1. The effect of YISHEN decoction on the renal function of rat models with 5/6 nephrectomy .Untill 8 weeks after 5/6 nepyrectomy ,rats both in model group and therapy group suffer with serious damage in renal function .We can see that the rats of both groups have high level of serum BUN and Cr than normal group. But rats of therapy group have more heavy mean avoirdupois, higher livability,longer lives, so they have high quality of life. However, we did't find significant difference on the level of serum BUN or Cr between model group and therapy group. Thus, we can't use YISHEN decoction as single method to improve renal function.2. The effect of YISHEN decoction on the pathologic changes of structure in remnant kidney of rats with 5/6 nephrectomy. Rats in model group have more cells in glomerulus than therapy group ,with more extracelluar matrix(ECM)and crescent shaping,and mesangial section is enlarged and dyed darkly,capillary vessel is open badly or even is obturated. Compared with those of the normal control group and the treatment group , the kidneys of the model group had more severe glomerulosclerosis, higher glomerulosclerosis index(GSI).While inflammation and hyperplasia cells are gentle evidencly in therapy group rats, and GSI is lower accordingly.3.The effect of YISHEN decoction on the expression of collagen IV(IV-col) and matrixmetalloproteinases 2(MMP-2).The degree of IV-col expression were allayed by YISHEN decoction treatment. In the experiment, with Immunohistochemistry method we find that there is more light dyeing and smaller stained area of IV-Col in remnant kidney of YISHEN therapy group rats .At the same time , the expression of MMP-2 is melody up with large areaand strong intensity.
