Objective:To observe the 24-hour urine protein quantification,blood biochemistry,renal histopathological changes,and transforming growth factor in renal tissue by observing the effect of Shenfukang VI intervention on focal segmental glomerulosclerotic adriamycin nephrotic syndrome rats.?1(TGF-?1),Matrix Metalloproteinase-2(MMP-2),Tissue Inhibitor of Metalloproteinase-2(TIMP-2),Collagen-IV(COL-IV)expression to investigate the renal protective effect of Shenfukang VI on focal segmental glomerulosclerosis in rats with adriamycin nephropathy and its possible mechanism of regulating renal fibrosis.Material Methods:Sixty male Sprague-Dawley rats were randomly divided into normal group,doxorubicin group,kidney rehabilitation group VI group,prednisone group,and combination group with 12 rats in each group.A rat model of focal segmental glomerulosclerosis with adriamycin nephropathy was established by repeated injection of adriamycin in the rat tail vein.After 6 weeks of modeling,the Renweikang VI group was given daily Renweikang VI decoction orally,prednisone group was given daily prednisone solution,and the combination group was given daily Renweikang VI decoction.Combined prednisone solution gavage,continuous gavage treatment for 4 weeks.Observing the general state of rats in each group,the 24-hour urinary protein quantitation,urine volume,and body weight were measured every weekend during the 0th to the 4th week of treatment,and serum total protein,albumin,globulin,blood lipid,and kidney function were measured at the 4th weekend of treatment.The histopathological changes of rat kidney were observed under light microscope.Immunohistochemical method was used to measure the expression of TGF-?1,MMP-2,TIMP-2 and COL-IV in rat kidney.SPSS 20.0 software was used to analyze the one-way ANOVA between groups.The data was expressed as x±s.The comparison between groups was performed using the LSD-t test.Results:1.Successful establishment of a rat model of focal segmental glomerulosclerosis adriamycin nephropathy,with a large amount of proteinuria(130.17 ± 25.02)mg · kg-1 · d,hypoalbuminemia(17.89 ± 2.92 g / L),hyperlipidemia(total cholesterol 7.70±2.97 mmol/L,triglyceride 7.81±3.52 mmol/L),edema symptoms,pathological changes under light microscopy and immunohistochemical expression consistent with focal segmental sclerosis nephropathy Syndrome performance.2.General status The general state of rats in the treatment group was significantly better than that in the adriamycin group.In the later period of the experiment,the weight of the rats in the combined group was higher than that in the doxorubicin group,and the difference was extremely statistically significant(P<0.01).3.At the end of the first week of the 24 h urinary protein dosing period,the 24-hour urinary protein quantification was significantly reduced in the dosing group compared with the doxorubicin group(P<0.01);the Kidney Recon VI side was combined with prednisone.Treatment of focal segmental glomerulosclerotic adriamycin-induced nephropathy in rats is more prominent than the single use of Renifukang VI decoction or prednisone.4.Serum total protein,albumin,and globulin were significantly increased in the treatment group compared with the adriamycin group.Among them,albumin was compared with doxorubicin group,kidney rehabilitation group VI(21.15±2.59g/L),prednisone group(20.68±2.60g/L),combined group(23.38±5.07g/L)Highly significant,the difference was statistically significant.5.Pathological changes of the kidneys The mesangial cells of the treated group showed mild hyperplasia,renal interstitial inflammatory cell infiltration was reduced,some interstitial fibrosis,occasional sclerosis lesions,and the lesions were significantly lighter than the model group.The expression of TGF-?1,TIMP-2 and COL-IV in the immunohistochemistry group was lower than that in the doxorubicin group,and the expression of MMP-2 was significantly increased.The expression of TGF-?1 and COL-IV in the combination group was lower than that in the kidney-rehabilitative VI group and prednisone group.The expression of TIMP-2was lower in the Renkangkang VI group and the combination group than in the prednisone group.There was no statistically significant difference in expression between treatment groups.Conclusion:This experimental study found that the therapeutic effect of Renfukang VI on rats with focal segmental sclerosis of adriamycin nephropathy: 1 Improves the general state of rats;2 Reduces 24 hours proteinuria,increases plasma total protein,white Protein,globulin;3to reduce renal pathological injury;4 kidney rehabilitation Kang side combined with prednisone treatment of adriamycin induced renal nephropathy than a single use of kidney rehabilitation VI decoction or prednisone more prominent.The therapeutic mechanism of Shenfukang VI may be through reducing the expression of inflammatory fibrogenic factor TGF-?1,regulating the matrix degrading enzyme system involved in MMP-2 and TIMP-2,and inhibiting the overexpression of extracellular matrix composed of COL-IV.Accumulation reduces rat urinary protein excretion,corrects hypoproteinemia,and reduces renal pathological damage.
