Association Of IL-1RN, TNF-A-308 Gene Polymorphisms With The Risk Of Gastric Cancer

Objective:To investigate the relationship between Interleukin-1RN, TNF-A-308 gene polymorphisms and the risk of gastric cancer in Anhui province, and find the difference of frequency of the genotypes of IL-1RN and TNF-A-308 between population with different races and different areas, and also analysis the association of these genotypes with H.pylori infection. Methods: Fifty-five health controls and ninty-one patients with gastric cancer in Anhui province were genotyped by polymerase chain reaction (PCR) for IL-1RN gene 86 bp variable number of tandem repeat (VNTR) polymorphism of IL-lRN gene intron 2(for five different alleles), and the TNF-A-308 polymorphisms were detacted by DNA sequencing. The H.pylori IgG antibody was detected by ELISA. RESULTS: (1)For IL-1RN gene polymorphism, we detected IL-1RN*1*1, IL-1RN*1*2, IL-1*2*2 and IL-1RN*1*3 four genotypes, the frequency of them in gastric cancer patients are 82.4%, 13.2%,2.2%, 2.2%, in controls are 83.7%, 12.7%, 1.8%, 1.8%, there was no significant diffierence of these genotypes between gastric cancer patients and controls(P>0.05). (2) Compared with population of different races and areas, we found that the frequency of genotypes of IL-1RN in our population was similar with Taiwan and other areas of Chinese mainland, but significant different from the European Caucasian population and Japanese population, especially in the IL-1RN*2*2 homozygote (P<0.05). (3) For TNF-A-308 gene polymorphism ,we detected two genotypes, it were TNF-A-308G/G homozygote and TNF-A-308G/A heterozygote, we have not detected the TNF-A-308A/A homozygote in our population, the frequency of these genotypes in gastric cancer patients are 87.9%, 12.1%, 0.0% , in controls are 87.3%, 12.7%, 0.0%, there was no significant difference of these genotypes between gastric cancer patients and controls(P>0.05). (4) Compared with population of different races and areas, we found the frequency of genotypes of TNF-A-308 was similar with other areas of Chinese mainland, But significantly different from that in Caucasian population(United states and Finland)and Japanese population especially in TNF-A-308G/A (P<0.05). (5) For H.pylori infection, we detected 53 positives in 91 patients (58.2%), and 29 positives in 55 controls (52.7%), there was no association of the frequency of genotypes of IL-1RN, TNF-A-308 with H.pylori infection.Conclusion: (1) IL-1RN, TNF-A-308 gene polymorphisms may be not associated with the risk of gastric cancer in this high-risk area. (2) There was a significant difference in the frequency of genotypes of IL-1RN and TNF-A-308 between different races and areas. (3)There was no association of IL-1RN, TNF-A-308 gene polymorphisms with H.pylori infection.
