Objective:To find wine-prepared Cornus officinalis quality marks,by studying the differences chemical compositions about wine-prepared Cornus officinalis and its raw.Established wine-prepared Cornus officinalis content determination and chromatographic fingerprint method to improve current quality control method.In order to provide reliably date support for the quality estimate of the medicines.Methods:(1)Refer to Chinese Pharmacopoeia of 2015,to establish an content determination method of loganin and morroniside by UPLC,for evaluate quality of 44 batches of wine-prepared Cornus officinalis and 55 batches of raw samples.(2)To establish an UPLC-MS method for identify chemical compositions of wine-prepared Cornus officinalis.(3)Basd on chemometrics,to recognition differences chemical compositions between wine-prepared Cornus officinalis and its raw.(4)To establish a PLS prediction model for distinguishing wine-prepared Cornus officinalis and its raw,and applying of 11 batches samples to verify this model.(5)To develop a quantitative analysis method of multi-component by UPLC,and compare their difference.(6)To establish UPLC fingerprint method of wine-prepared Cornus officinalis,and combine fingerprint similarity analysis with clustering analysis and principal component analysis to evaluate the medicine.Results:(l)The total contents of loganin and morroniside were below standard including 10 batches from raw Cornus officinalis,and 44 batches wine-prepared Cornus officinalis up to standard.(2)The total of 43 compounds were identified from wine-prepared Cornus officinalis,and 38 compounds were identified from the raw.(3)Chemometric analysis results indicate that screened and obtained 24 components which have a great difference.(4)The PLS prediction model can predict unknown sample,accurately.(5)To develop a quantitative analysis method of 10 components by UPLC,method validation showed that liner relationship and precision,repeatability and stablility,the recoveries are meet with Chinese Pharmacopoeia.(6)The UPLC fingerprints of wine-prepared Cornus officinalis were established,and 18 common peaks of fingerprints of wine-prepared samples were identified.The similarity analysis of 45 samples showed that the similarities were above 0.8with 32,and below 0.8 with 13.The UPLC fingerprints of raw Cornus officinalis were established,and 15 common peaks of fingerprints were identified.The similarity analysis of 45 samples showed that the similarities were above 0.8 with 37,and only 7 batches below 0.8.The quality of the raw samples are relatively stable from different rigions,and the wine-prepared samples are significantly different.Conclusion:Based on above results,UPLC-MS can be used to provide an effective way for medicine informations.The UPLC quantitative analysis and fingerprint research methods of Cornus officinalis were initially established,the results of stability,precision and repeatability were meet with the requirements,and could be used widely in Cornus officinalis of quality evaluation.
