| Objective:To figure out the correlations among a single nucleotide polymorp hism site rs2036914,plasma level of FⅪ and pulmonary thromboembolism in t he Shenzhen area of southern China by analyzing the SNP and plasma concent rations of FⅪ.Methods:126 newly diagnosed unrelated sporadic PTE patients from May 2017 to September 2019 in Shenzhen People’s Hospital were selected in the case group,and 99healthy subjects were in the control group.Clinical basic data and morning fasting blood of the subjects were collected.Use Elisa to detect the plasma concentration of FⅪ.Use Sanger sequencing to detect and identify the genotype and alleles of FⅪ rs2036914.Statistically analyze the relationship among rs2036914,plasma level of FⅪ and pulmonary thromboembolism.Result:1.The amount of genotypes CC,CT,and TT of FⅪ rs2036914 were 80(63.5%),40(31.7%),and 6(4.8%)in the PTE group,and 46(46.5%),47(47.5%),and 6(6.1%)in the control group(P=0.037).The frequency of C allele(79.4%)in the PTE group was higher than that in the control one(71.2%),which increased the risk of PTE(P=0.025,ORC=1.663 95%CI(1.061-2.512))).Univariate regression analysis shows that in the recessive model,the CC genotype increased the risk of PTE compared with the CT+TT genotype.(P=0.011,ORCC=2.004,95%CI(1.172-3.245))and in the dominant model,there was no significant difference between the two groups.2.There were no significant differences between rs2036914 and the severity of PTE.3.The plasma FⅪ level in the PTE group(1286.95±255.04ng/ml)was higher than that in the control group(1196.04±317.09ng/ml).There was no correlation between FⅪ concentration and the severity of PTE.4.There were statistically significant differences in FⅪ level among the CC,CT,and TT genotypes of FⅪ rs2036914 in the PTE group,but no significant difference in the control group.In the PTE group,the plasma FⅪ concentration of genotype CC(1334.94±271.27ng/ml)was significantly higher than that of genotype CT(1211.70±198.70ng/ml),TT(1106.57±255.36ng/ml).There was no statistically difference between the plasma FⅪ of genotype CT and TT.5.The FⅪ rs2036914CC genotype(non-T allele carriers)(OR=1.913,95%CI1.091~3.356)and hypertension(OR=1.918,95%CI 1.030~3.573)are two independent risk factors for PTE(P<0.05),while smoking and FⅪ level weren’t.Conclusion:1.FⅪ rs2036914 gene polymorphism is closely related to the risk of pul monary thromboembolism,which may be a susceptible gene locus for pulmonar y thromboembolism.2.The plasma level of FⅪ elevates in patients with pulmonary thromboe mbolism.Plasma level of FⅪ elevates in non-T allele carriers.3.It has not yet been proven that rs2036914 and the plasma level of F Ⅺ are related to the severity of PTE.4.FⅪ rs2036914CC genotype is the independent risk factor for PTE. |
